摘  要:目的 基于自噬相关蛋白探讨天门冬提取物对人结肠癌细胞SW620的作用机制。方法 采用噻唑蓝(MTT)法、平板克隆形成实验、EdU/Hoechst33258荧光染色检测苗药天门冬提取物对SW620细胞增殖的影响,通过流式细胞术检测细胞凋亡率,Western印迹检测自噬及凋亡相关蛋白表达。结果 天门冬对SW620有显著的生长抑制及促凋亡作用,且具有明显剂量依赖性(P<0.05)。MTT、平板克隆形成实验、EdU/Hoechst33258荧光染色法实验结果提示,天门冬可明显抑制SW620细胞增殖(P<0.05)。Western印迹结果提示,天门冬各浓度组B细胞淋巴瘤(Bcl)-2相关X蛋白(Bax)、自噬相关蛋白(Beclin)1及微管相关蛋白轻链(LC)3-Ⅱ/LC3-Ⅰ蛋白表达明显高于空白对照组,Bcl-2和P62蛋白表达明显低于空白对照组(P<0.05)。结论 天门冬提取物可能是通过自噬途径促进SW620细胞凋亡,进而抑制结直肠癌细胞增殖。Objective To investigate the effect of asparagus extract on human colon cancer cells SW620 based on autophagy-re-lated proteins.Methods 3-(4,5)-dimethylthiahiazo(-z-y1)-3,5-di-phenytetrazoliumromide(MTT)method,plate clone formation as-say,EdU/Hoechst 33258 fluorescence staining were used to detect the effect of Miao medicine asparagus extract on the proliferation of SW620 cells,apoptosis rate was detected by flow cytometry and expressions of autophagy and apoptosis-related protein were detected by Western blot.Results The growth was significanthy inhibited and apopotosis was significantly promoted in SW620 by asparagus with a significant dose-dependent manner(P<0.05).The results of a MTT,colony formation test,EdU/Hoechst33258 fluorescence staining re-vealed that cell proliferation was significantly inhibited by asparagus(P<0.05).Western blot results showed that the expressions of B-cell lymphoma(Bcl)-2 associated X protein(Bax),autophagy associated proteins(Beclin)1 and microtubule-associated protein light chain(LC)3-Ⅱ/ LC3-Ⅰprotein were significantly higher in the asparagus treatment group than those in the blank control group, and the expres-sions of Bcl-2 and P62 protein were significantly lower than those in the blank control group(P<0. 05). Conclusions Asparagus extractmight promote apoptosis of SW620 cells through the autophagic pathway, thereby inhibiting the proliferation of colorectal cancer cells.
