摘 要:目的利用高通量测序技术进行全转录测序,找寻慢性阻塞性肺疾病(COPD)相关肺动脉高压(PH)患者中差异性表达的mRNA、lncRNA,并对差异表达的mRNA、lncRNA进行生物信息学分析,以期为COPD相关PH的靶向治疗提供依据并寻找潜在靶点。方法采用高通量测序技术对COPD相关PH患者、COPD患者及正常人(每组各6例)外周血细胞进行全转录测序,基于比对结果,进行基因表达量及差异性表达分析。结果识别差异表达基因2060个,COPD相关PH组vs COPD组共有805个上调的mRNA和297个下调的mRNA,COPD相关PH组vs正常对照组共有780个上调mRNA和355个下调mRNA。COPD相关PH组与COPD组、正常对照组分别进行比较,共有1886个差异表达lncRNA,其中1115个lncRNA差异表达上调,771个差异表达下调。差异表达的基因主要集中在感染、免疫、炎症、细胞核因子κB、肿瘤坏死因子信号通路等方面。结论COPD相关PH患者外周血中mRNA、lncRNA存在差异性表达,这些差异表达的基因可能参与COPD相关PH的发生发展过程,可能为COPD相关PH的早期诊断及靶向治疗提供新的切入点。Objective To find differentially expressed mRNA and lncRNA in patients with pulmonary hypertension(PH)as-sociated with chronic obstructive pulmonary disease(COPD),and analyze bioinformatics for mRNA and lncRNA with differential ex-pressions by conducting total transcription sequencing with high throughput sequencing technology,in order to provide basics for targe-ted therapy of COPD related PH and find potential targets.Methods High throughput sequencing technology was used to perform full transcriptional sequencing on peripheral blood cells of patients with COPD related PH,COPD patients and normal subjects(6 cases in each group).Based on the comparison results,gene expression levels and differential expressions were analyzed.Results 2060 dif-ferentially expressed genes were identified.Compared with COPD group,there were 805 mRNA up-regulated and 297 mRNA down-regulated in COPD related PH group.Compared with normal control group,there were 780 mRNA up-regulated and 355 mRNA down-regulated in COPD related PH group.Compared with COPD group and the normal control group,there were 1886 lncRNA differential-ly expressed in COPD related PH group,1115 lncRNA were up-regulated and 771 lncRNA were down-regulated.Differentially ex-pressed genes were mainly concentrated in infection,immunity,inflammation,nuclear factorκB and tumor necrosis factor signaling pathways.Conclusions mRNA and lncRNA in peripheral blood of patients with COPD related PH are differentially expressed,and these mRNA and lncRNA may be involved in the occurrence and development of COPD related PH,which may provide a new point for the early diagnosis and targeted therapy of COPD related PH.
