摘  要:目的研究衰老对小鼠角膜中配对盒基因(Pax)6和Wnt7a表达的影响,探索其与角膜缘干细胞(LSCs)衰老的关系。方法60只雌性昆明小鼠分为3组:青年组(2月龄),中年组(10月龄),老年组(18月龄)各20只。苏木素-伊红(HE)染色观察角膜形态结构改变,免疫荧光染色和流式细胞术检测p63、Pax6和Wnt7a分布和表达,实时荧光定量-聚合酶链反应(RT-qPCR)检测p63、Pax6和Wnt7a mRNA相对表达。结果与青年组相比,随着月龄的增加,小鼠角膜上皮的厚度明显变薄,角膜厚度明显增厚,p63、Pax6和Wnt7a表达均逐渐明显减少(P<0.05)。结论随着增龄,角膜呈老化趋势,可能与Pax6-Wnt7a调控有关。Objective To explore the effect of aging on expressions of paired box gene(Pax)6 and Wnt7a in mouse cornea,as well as the relationship with the aging of limbal stem cells(LSCs).Methods Sixty female kunming mice were divided into three groups:youth group(2 months),middle age group(10 months)and old age group(18 months),with 20 rats in each group.The structure of mouse cornea section was observed by hematoxylin-eosin(HE)staining.The distribution and expressions of p63,Pax6 and Wnt7a were detected by immunofluorescence staining and flow cytometry.The mRNA relative expressions of p63,Pax6 and Wnt7a were detected by quantitative real-time polymerase chain reaction(RT-qPCR).Results Compared with youth group,corneal epithelium thickness was significantly thinner and corneal thickness was significantly increased,and the expressions of p63,Pax6 and Wnt7a were gradually and significantly decreased with the increase of age(P<0.05).Conclusions With the increase of age,cornea could show an aging trend,which might be related to the regulation of Pax6-Wnt7a.
