摘  要:长链脂酰辅酶A合成酶(long-chain acyl-coenzyme A synthetase,LACS)是酰基激活酶(acyl-activating enzyme,AAE)超家族的重要成员,在植物应答生物和非生物胁迫时发挥重要作用。蒿叶猪毛菜具有较强的抗旱性。为探究LACS基因在蒿叶猪毛菜抗旱性中的生物学功能,本研究以蒿叶猪毛菜幼苗为材料,利用qRT-PCR技术对蒿叶猪毛菜中的SaLACS1基因进行克隆,并通过生物信息学方法分析基因表达模式,再对干旱胁迫下SaLACS1基因进行转录组表达分析及qRT-PCR验证。结果表明:(1)克隆到的SaLACS1基因,编码区长度大小为1641 bp,可编码546个氨基酸。(2)生物信息学分析表明,该蛋白定位于过氧化物酶体且为稳定蛋白,主要由α螺旋和无规则卷曲组成。(3)系统发育树分析表明,SaLACS1蛋白与甜菜、菠菜同源蛋白具有更近的亲缘关系。(4)SaLACS1启动子预测分析发现,该启动子区存在多种与植物生长发育、胁迫响应相关的顺式作用元件。(5)qRT-PCR结果表明,在干旱胁迫下,SaLACS1基因上调,表达量显著增加。综上所述,SaLACS1基因在蒿叶猪毛菜抗旱过程中可能参与了与干旱胁迫响应相关的调控机制,为后续探究蒿叶猪毛菜SaLACS1基因功能提供理论依据。Long-chain acyl-coenzyme A synthase(LACS)is an important member of acyl activating enzyme(AAE)superfamily,playing a crucial role in plant responses to biotic and abiotic stress.Salsola abrotanoides L.has strong drought resistance.In order to explore the biological function of LACS gene in the drought resistance of Salsola abrotanoides L.,its seedlings are taken as materials to clone SaLACS gene using qRT-PCR technology.The gene expression pattern is analyzed by bioinformatics.The transcriptome expression of SaLACS gene under drought stress is analyzed and verified by qRT PCR.The results are as follows:(1)The cloned SaLACS1 gene has a coding region length of 1641 bp and can encode 546 amino acids.(2)Bioinformatics analysis shows that its protein is located in peroxisome and is a stable protein,mainly composed of alpha helices and irregular coils.(3)Phylogenetic tree analysis shows that SaLACS1 protein has a closer phylogenetic relationship with homologous proteins from sugar beet and spinach.(4)The prediction analysis of SaLACS1 promoter reveals that there are multiple cis acting elements related to plant growth,development and stress response in this promoter region.(5)The results of qRT-PCR shows that SaLACS1 gene is up-regulated and its expression level significantly increases under drought stress.In summary,SaLACS1 gene may be involved in the regulation mechanism related to drought stress response in the drought resistance process of Salsola abrotanoides L.,providing a theoretical basis for further research on the function of SaLACS1 gene in Salsola abrotanoides L.
