摘  要:目的:建立HPLC-CAD法同时测定二冬膏中麦冬皂苷D、麦冬皂苷D′、菝葜皂苷元和薯蓣皂苷4种成分含量的方法。方法:色谱柱为InfinityLab Poroshell 120 Aq-C_(18)色谱柱(150 mm×3.0 mm,2.7μm),流动相乙腈-0.1%甲酸水溶液梯度洗脱,柱温35℃,流速1 ml/min,进样量10μl。电喷雾检测器的雾化器温度为35℃,检测频率为10 Hz。结果:麦冬皂苷D、麦冬皂苷D′、菝葜皂苷元和薯蓣皂苷分别在20~300、10~150、30~450和60~900μg/ml浓度范围内与峰面积线性关系良好(r=0.9996、0.9998、0.9998、0.9997),麦冬皂苷D、麦冬皂苷D′、菝葜皂苷元和薯蓣皂苷平均加样回收率分别为98.5%(RSD为0.5%)、98.9%(RSD为1.1%)、99.1%(RSD为1.2%)和100.4%(RSD为0.9%)。结论:该方法可用于二冬膏中麦冬皂苷D、麦冬皂苷D′、菝葜皂苷元和薯蓣皂苷的含量测定。Objective:To establish an HPLC-CAD method for the simultaneous determination of the contents of four components:ophiopogonin D,ophiopogonin D′,sarsasapogenin,and dioscin in Erdong Gao.Methods:The chromatographic column was an InfinityLab Poroshell 120 Aq-C_(18)(150 mm×3.0 mm,2.7μm),with a gradient elution of acetonitrile(A)-0.1%formic acid aqueous solution(B)as the mobile phase.The column temperature was 35℃,flow rate was 1 ml/min,and injection volume was 10μl.The nebulizer temperature of the electrospray detector was 35℃and the detection frequency was 10 Hz.Results:There was a good linear relationship between the peak area and the concentrations of ophiopogonin D,ophiopogonin D′,sarsasapogenin,and dioscin in the ranges of 20~300,10~150,30~450,and 60~900μg/ml,respectively(r=0.9996,0.9998,0.9998,0.9997).The average recovery rates of ophiopogonin D,ophiopogonin D′,sarsasapogenin,and dioscin were 98.5%(RSD of 0.5%),98.9%(RSD of 1.1%),99.1%(RSD of 1.2%),and 100.4%(RSD of 0.9%),respectively.Conclusion:This method can be used for the determination of ophiopogonin D,ophiopogonin D′,sarsasapogenin,and dioscin in Erdong Gao.
