摘  要:目的建立LPS诱导的小鼠单核巨噬细胞(RAW264.7)炎症模型,探究Rhamnazin的抗炎活性及其作用机制。方法CCK-8法测定Rhamnazin对单核巨噬细胞活力的影响,ELISA法测定细胞上清液中小鼠白介素6(IL-6)、白介素1β(IL-1β)及肿瘤坏死因子α(TNF-α)的含量及其mRNA表达水平,Western blot法检测NF-κB、p-NF-κB、ERK、p-ERK、JNK、p-JNK、p38MAPK和p-p38MAPK蛋白的表达水平。结果Rhamnazin可明显抑制LPS诱导的RAW264.7细胞培养液中炎症因子的表达水平;抑制NF-κB、p38、ERK和JNK蛋白磷酸化水平。结论Rhamnazin可能通过抑制TLR4/IκB/NF-κB信号通路,抑制巨噬细胞LPS诱导的炎性反应中IL-6、IL-1β和TNF-α等炎症因子的释放,并通过抑制NO释放、XOD活性和清除DPPH自由基发挥抗氧化活性和抗炎活性。Objective To establish an LPS-induced model of mononuclear macrophage(RAW264.7)inflammation in mice,and explore the anti-inflammatory activity and mechanism of Rhamnazin.Methods CCK-8 method was used for determination of the impact of Rhamnazin on cell viability;Elisa method was used to determine the contents of interleukin 6(IL-6),interleukin 1β(IL-1β)and tumor necrosis factorα(TNF-α)and their mRNA expression levels in the cell supernatant.The Western blot method was used to detect the expression levels of NF-κB,p-NF-κB,ERK,p-ERK,JNK,p-JNK,p38MAPK,and p-p38MAPK proteins.Results Rhamnazin had a significant inhibitory effect on the secretion of inflammatory factors in LPS-induced RAW264.7 cell culture solution and inhibited the phosphorylation levels of NF-κB,p38,ERK and JNK proteins.Conclusions Rhamnazin may exert anti-inflammatory activity by inhibiting the TLR4/IκB-α/NF-κB signaling pathway and inhibiting the release of inflammatory factors such as IL-6,IL-1βand TNF-α.It also exerted antioxidant and anti-inflammatory activities by inhibiting NO release,XOD activity and scavenging DPPH free radicals.
