摘 要:为鉴定发生在哈尔滨市旱柳Salix matsudana上的一种锈病病原,采用光学和扫描电子显微镜观察其夏孢子形态学特征,提取其夏孢子基因组DNA,扩增ITS和LSU基因并测序,依据ITS和LSU串联序列构建系统发育树。系统发育树上该病原菌与铁锈栅锈菌Melampsora ferrinii汇为一支,结合形态学鉴定,确定该病原菌是铁锈栅锈菌Melampsora ferrinii Toome&Aime。该锈菌是东北地区的锈菌新记录种,旱柳是该锈菌在国际首次报道的新寄主植物。To identify the pathogen of rust disease on Salix matsudana,the morphological characteristics of urediospores were observed using both light microscope and scanning electron microscope.The genomic DNA of urediospores were extracted,PCR amplification was performed on the ITS and LSU of ribosomes to further construct a phylogenetic tree.The pathogen was found to cluster with Melampsora ferrinii on the phylogenetic tree.Combined with morphological identification,the pathogen was determined to be M.ferrinii Toome&Aime.M.ferrinii Toome&Aime was a new record species in the Northeast region of China,and S.matsudana was reported as a new host plant for this fungus internationally for the first time.
