福建省薯蓣2种病原线虫的种类形态与分子鉴定

摘  要:[目的]明确福建省薯蓣(Dioscorea opposite)病原线虫种类,为其科学防控提供依据。[方法]通过样本采集及症状观察,利用线虫形态学观测和分子生物学检测方法鉴定线虫种类。[结果]古田县薯蓣样本呈根结线虫为害症状,通过线虫形态特征观测,并结合rDNA-ITS区、28S D2-D3区、mtDNA(COII/lrRNA)序列分析及特异性引物检测,确定为南方根结线虫(Meloidogyne incognita);永安县薯蓣样本呈短体线虫为害症状,通过形态学鉴定,并结合rDNA-ITS区序列分析和特异性引物检测,确定为咖啡短体线虫(Pratylenchus coffeae)。[结论]首次报道了南方根结线虫与咖啡短体线虫为福建省薯蓣重要的病原线虫,选用健康薯蓣并采取水旱轮作以减少初侵染源是重要的防控手段。[Purpose]The aim of this study was to identify the species of pathogenic nematodes of Dioscorea opposita in Fujian Province and provide the basis for formulating effective prevention and control measures for nematode diseases.[Method]Samples were collected,symptoms were observed,and the nematode species were identified based on morphological and molecular characteristics.[Result]The samples of D.opposita in Gutian County showed the symptoms of Meloidogyne spp.infestation.Based on the morphological characteristics;sequence analysis of the ribosomal DNA internal transcribed spacer(ITS)region,28S D2-D3 region and mitochondrial DNA(COII/lrRNA);and specific primer detection,the nematode was identified as M.incognita.The samples of D.opposita in Yong′an County showed the symptoms of Pratylenchus spp.infestation.Based on morphological characteristics,sequence analysis of DNA ITS region,and specific primer detection,the nematode was identified as P.coffeae.[Conclusion]For the first time,we report that M.incognita and P.coffeae are important causes of D.opposite nematode diseases in Fujian Province.Selecting healthy seed tubers or sections of tuber and taking paddy-upland measures to reduce the sources of primary infection are important prevention and control measures.
