槲皮素对副猪嗜血杆菌感染猪气管上皮细胞RPSA、MAPK/NF-κB信号通路的调控作用

摘  要:本试验旨在研究槲皮素对副猪嗜血杆菌(GPS)感染的新生猪气管上皮细胞(NPTr细胞)RPSA、MAPK/NF-κB信号通路的影响。试验采用GPS感染NPTr细胞作为细胞屏障损伤模型,采用Western blot检测NPTr细胞中RPSA、MAPK/NF-κB信号通路蛋白表达水平,通过设计RPSA干扰引物和RPSA过表达载体,进一步研究槲皮素对GPS感染的NPTr细胞RPSA、MAPK/NF-κB通路的影响。结果显示,与空白对照组相比,GPS感染NPTr细胞后RPSA蛋白、JNK、ERK、p38和NF-κB磷酸化蛋白表达水平均显著上调(P<0.05或P<0.01);与GPS感染组相比,槲皮素的预处理可下调NF-κB、JNK、ERK和p38蛋白的磷酸化水平;RPSA siRNA能够干扰RPSA的表达并降低MAPK/NF-κB相关通路蛋白的表达;RPSA的过表达可显著上调NF-κB、JNK、ERK、p38磷酸化蛋白表达水平;槲皮素处理能显著降低RPSA相关信号通路蛋白的表达水平(P<0.05或P<0.01)。结果提示,槲皮素能够通过调控RPSA、MAPK/NF-κB信号通路来缓解GPS感染所致的气管上皮细胞损伤。The aim of this experiment was to investigate the effects of quercetin on the RPSA and MAPK/NF-κB signalling pathways in Haemophilus parvovirus(GPS)-infected neonatal porcine tracheal epithelial(NPTr)cells.GPSinfiltrated NPTr cells were used as a model of cellular barrier damage,and Western blot was used to detect the expression levels of RPSA protein and MAPK/NF-κB signalling pathways protein.The effects of quercetin on RPSA,MAPK/NF-κB pathways in GPS-infected NPTr cells were further investigated by designing RPSA-interfering primers and overexpressing RPSA vectors.The results showed that the expression levels of RPSA proteins,JNK,ERK,p38 and NF-κB phosphorylated proteins were significantly upregulated after GPS infection of NPTr cells compared to the control group(P<0.05 or P<0.01).Compared to the GPS-infected group,pretreatment with quercetin could reduce the phosphorylation levels of NF-κB,JNK,ERK and p38 proteins.RPSA siRNA could interfere with the expression of RPSA and reduce the expression of MAPK/NFκB-related pathway proteins.RPSA overexpression could significantly increase the levels of NF-κB,JNK,ERK and p38 phosphorylated proteins,and quercetin treatment could significantly decrease the expression of RPSA-related signalling pathway proteins(P<0.05 or P<0.01).The results indicate that quercetin can alleviate GPS infection-induced tracheal epithelial cell injury by modulating RPSA and MAPK/NF-κB signalling pathways.
