摘  要:目的观察快速老化(senescence-accelerated mouse prone 8,SAMP8)小鼠的肝脏炎性反应,探讨电针通过调控肝脏组织中糖原合成酶激酶-3β(glycogen synthase kinase-3β,GSK-3β)/雷帕霉素机械靶蛋白(mechanistic target of rapamycin,mTOR)通路对SAMP8小鼠肝脏炎性反应的影响。方法选用7月龄的SAMP8小鼠及同龄且遗传背景相同的抗快速老化(senescence-accelerated mouse resistant 1,SAMR1)小鼠作为实验对象。随机将SAMP8小鼠分为电针组和模型组,每组12只;正常组为12只SAMR1小鼠。电针组分别在“百会”穴和双侧“肾俞”穴进行针刺,电针双侧“肾俞”穴。在相同时间,正常组和模型组进行与电针组一样的抓取束缚,不作任何干预。干预后,观察3组小鼠一般情况,采用免疫印迹法(Western blot)检测3组小鼠肝组织GSK-3β、mTOR、白介素-6(interleukin-6,IL-6)、白介素-4(interleukin-4,IL-4)蛋白表达,采用苏木素-伊红(hematoxylin-eosin staining,HE)染色观察肝组织病理改变。结果模型组肝组织GSK-3β、mTOR、IL-6蛋白表达水平均高于正常组(P<0.05),IL-4蛋白表达水平低于正常组(P<0.05),肝组织炎性细胞浸润程度较严重。电针组肝组织GSK-3β、mTOR、IL-6表达均低于模型组(P<0.05),IL-4蛋白表达高于模型组(P<0.05),肝组织炎性细胞浸润程度较模型组减轻。结论电针可能通过抑制肝脏组织中GSK3β/mTOR促炎通路的表达,降低肝脏组织中的炎性反应。Objective By observing the inflammatory responses in the liver of senescence-accelerated mouse prone 8(SAMP8)mice,to investigate the effect of electroacupuncture(EA)on inflammatory responses by regulating the glycogen signaling pathway synthase kinase-3β(GSK-3β)/mechanistic target of rapamycin(mTOR)in the liver tissues.Method Seven-month-old SAMP8 mice and senescence-accelerated mouse resistant 1(SAMR1)mice of the same age and genetic background were taken as the subjects.The SAMP8 mice were randomized into an EA group and a model group,with 12 mice in each group;12 SAMR1 mice were taken as the normal group.Mice in the EA group received acupuncture at“Baihui(GB20)”and bilateral“Shenshu(BL23)”,with bilateral“Shenshu”connected to EA.Simultaneously,the normal and model groups received the same grasping and binding as the EA group without any intervention.When the intervention finished,the three groups of mice were observed for their general condition;Western blot was used to detect the protein expression of GSK-3β,mTOR,interleukin-6(IL-6),and interleukin-4(IL-4);the histopathological changes of liver tissues were observed using hematoxylin-eosin(HE)staining.Result Compared to the normal group,the protein expression levels of GSK-3β,mTOR,and IL-6 in the liver tissues were higher in the model group(P<0.05),and the protein expression level of IL-4 was lower in the model group(P<0.05);the liver tissues showed more severe inflammatory cell infiltration in the model group.Compared to the model group,the expression levels of GSK-3β,mTOR,and IL-6 in the liver tissues were lower in the EA group(P<0.05),and the protein expression level of IL-4 was higher in the EA group(P<0.05);inflammatory cell infiltration in the liver tissues improved in the EA group compared to the model group.Conclusion EA may reduce inflammatory responses in the liver tissues by inhibiting the expression of the proinflammatory signaling pathway GSK3β/mTOR in the liver.
