摘 要:【目的】旨在选出高多态性、高稳定性、可重复性好的核心引物,并找到更适合筛选甜菜CBDP引物扩增的PCR程序。【方法】利用3种不同的PCR程序,对39条CBDP引物进行筛选,并通过此3种PCR程序的扩增结果进行比较分析。【结果】Touch Down程序扩增结果优于固定退火温度55℃以及低退火温度的程序,基于最优退火温度从39条CBDP引物中筛选出9条多态性好的CBDP引物,这9条引物扩增出的总条带数为76条,多态性条带为72条,多态百分比为94.7%。多态性信息量范围在0.552~0.913,引物的平均PIC值为0.762。【结论】CBDP核心引物的筛选为进一步研究甜菜种质资源的遗传多样性提供了新的思路,为利用分子标记辅助育种等相关研究奠定了基础。【Objective】This paper aims to select core primers with high polymorphism,high stability and reproducibility,and find the most suitable PCR program for amplifying beet CBDP primers.【Methods】Using three different PCR programs to screen 39 CBDP primers,the amplification results of the three PCR programs were compared and analyzed.【Results】The amplification results of the Touch Down program were better than the fixed annealing temperature of 55℃and low annealing temperature.Based on the optimal annealing temperature,9 CBDP primers with good polymorphism were selected from 39 CBDP primers.The total number of bands amplified by 9 primers was 76,polymorphic bands was 72,and the polymorphic percentage was 94.7%.The polymorphism information content(PIC)ranged from 0.552 to 0.913,with an average PIC of the primers was 0.762.【Conclusion】The screening of CBDP core primers provides a new idea for further research on the genetic diversity of sugar beet germplasm resources,and lays a foundation for the use of molecular marker-assisted breeding.
