摘 要:目的通过研究五味子乙素(Schisandrin B,SchB)对痛风模型大鼠的影响,探讨其对痛风性肾损伤的保护作用及机制。方法采用腺嘌呤和氧嗪酸钾混合剂灌胃造模,酶联免疫吸附法检测血清中TNF-α、IL-1β及IL-6水平,试剂盒检测血清中BUA、Cr、BUN、XOD、SOD、MDA含量及尿液中UUA含量;HE染色观察肾脏病理学改变;Western blot法检测肾组织中TLR4、MyD88、NF-κB蛋白表达水平。结果经SchB治疗后血清中TNF-α、IL-1β、IL-6、BUA、Cr、BUN含量降低,XODase酶活性显著降低,尿液中UUA含量增加,SOD活力提高,MDA含量降低;肾脏病理学病变减轻;肾组织中TLR4、MyD88、NF-κB蛋白表达显著降低。结论SchB能改善腺嘌呤和氧嗪酸钾混合剂引起的大鼠痛风性肾损伤,减轻炎症,减少氧化应激损伤,减少尿酸生成,促进尿酸排泄,对大鼠痛风性肾损伤具有保护作用。To investigate the protective effect and mechanism of Schisandrin B(SchB)against gouty renal injury by studying its effects in gout model rats.Methods A mixture of adenine and potassium oxonate was used for gavage modeling.An enzyme-linked immunosorbent assay(ELISA)was employed to detect the levels of TNF-α,IL-1β,and IL-6 in serum.A kit was used to measure the levels of BUA,Cr,BUN,XOD,SOD,and MDA in serum,as well as the levels of UUA in urine.Hematoxylin and eosin(HE)staining was utilized to observe renal pathological changes,and Western blot analysis was conducted to assess the protein expression levels of TLR4,MyD88,and NF-κB in renal tissues.Results The serum levels of TNF-α,IL-1β,IL-6,BUA,Cr,and BUN were reduced after SchB treatment.Additionally,XODase activity was significantly reduced,urinary UUA content was increased,SOD viability was improved,and MDA content was reduced.Renal pathological lesions were also reduced,and the protein expression of TLR4,MyD88,and NF-κB in renal tissues was significantly decreased.Conclusion SchB ameliorates gouty renal injury induced by a mixture of adenine and potassium oxonate in rats.It reduces inflammation,oxidative stress injury,and uric acid production,promotes uric acid excretion,and provides protective effects against gouty renal injury.
