山油柑枝叶中化学成分及其抗类风湿性关节炎活性研究

摘  要:目的研究芸香科山油柑属植物山油柑Acronychia pedunculata枝叶中的化学成分。方法综合运用多种色谱分离方法和技术进行系统分离和纯化,根据分离得到化合物的理化性质及其NMR波谱数据和MS数据鉴定化合物的化学结构。结果从山油柑枝叶的提取物中分离得到了16个化合物,分别鉴定为阿里二醇(1)、款冬二醇(2)、马尼拉二醇(3)、齐墩果-12-烯-3,11-二酮(4)、16β-羟基-羽扇豆-20(29)-烯-3-酮(5)、岩高兰三萜-21-烯-3-酮(6)、24-亚甲基环阿尔廷醇(7)、5,6,7-三甲氧基香豆素(8)、6-羟基-7,8-二甲氧基香豆素(9)、6,7,8-三甲氧基香豆素(10)、秦皮素(11)、丝立尼亭(12)、紫花前胡苷元(13)、佛手酚(14)、佛手柑内酯(15)和花椒毒素(16)。其中化合物1~7为三萜类化合物,化合物8~16为香豆素类化合物。对分离鉴定的化合物1~16的抑制滑膜成纤维细胞增殖活性研究评价结果表明,化合物1~7对MH7A滑膜成纤维细胞增殖抑制活性的半数抑制浓度(median inhibition concentration,IC50)值为(9.26±0.10)~(156.28±0.17)μmol/L。结论所有化合物均为首次从山油柑属植物中分离得到,化合物1和4表现出较为显著的抗类风湿性关节炎活性。Objective To study the chemical constituents from the stems and leaves of Acronychia pedunculata.Methods The chemical constituents of A.pedunculata were isolated and purified by a variety of chromatographic methods and techniques including silica gel,ODS,Sephadex LH-20 gel column chromatographies as well as preparative HPLC.Their chemical structures were identified according to their physicochemical properties,NMR spectral data,MS data,as well as comparisons with the reported data in literature.Results A total of 16 compounds were isolated from the 90%ethanol extract of the stems and leaves of A.pedunculata,which were identified as arnidiol(1),faradiol(2),maniladiol(3),olean-12-ene-3,11-dione(4),16β-hydroxylupa-20(29)-en-3-one(5),nigrum-21-en-3-one(6),24-methylenecycoartanol(7),5,6,7-trimethoxy coumarin(8),6-hydroxy-7,8-dimethoxy-coumarin(9),6,7,8-trimethoxycoumarin(10),fraxidin(11),selinidin(12),nodakenetin(13),bergaptol(14),bergapten(15)and xanthotoxin(16).All isolated compounds were 1-16 were evaluated for their anti-rheumatoid arthritis activities using MTT methods,as a result,compounds 1-7 exhibited the inhibitory effects on the proliferation of MH7A synovial fibroblast cells with the IC50 values in range of(9.26±0.10)μmol/L-(156.28±0.17)μmol/L.Conclusion All compounds were isolated from the genus Artocarpus for the first time,compounds 1-7 showed remarkable anti-rheumatoid arthritis activities.
