摘 要:目的检测宁夏本土道地药材密点麻蜥单体成分代谢产物,筛选其相关代谢成分中与治疗疾病有关的代谢物质。方法运用T3方法色谱采集条件色谱质谱检测、非靶向代谢组技术检测人工养殖密点麻蜥(干品)混样,通过高效液相色谱(ultra performance liquid chromatography,UPLC)和串联质谱(tandem mass spectrometry,MS/MS)技术,将鉴定出样本中的代谢物与数据库结合进行代谢物结果分析。结果将密点麻蜥单体醇提液质谱图与各物质对照品质谱图进行比对,按照物质二级分类,分别鉴定出有机酸及其衍生物、氨基酸及其衍生物、小肽、氧化脂质、核甘酸及其代谢物、酰基肉碱、氨基酸、溶血磷脂酰胆碱、游离脂肪酸、溶血磷脂酰乙醇胺、胺类、苯及其衍生物、辅酶和维生素、酚酸类、糖类、吲哚及其衍生物、多胺、蝶啶及其衍生物、激素及激素相关物质、酯类、单甘油酯、胆汁酸、苯酚类、糖醇、醇类、其他、糖酸、磷酸糖、杂环化合物、磺酸类化合物、酮类、胆碱类、磷酸类化合物、糖衍生物、鞘磷脂、醛类、色胺、烃衍生物、甘油二酯、磷脂酰胆碱、磷脂酰肌醇、磷脂酰乙醇胺、色素等43类二级物质,其中排前三为有机酸及其衍生物(108种)、氨基酸及其衍生物(98种)、小肽(83种)、氧化脂质(83种)。结论密点麻蜥代谢物氨基酸及其衍生物、有机酸及其衍生物、小肽及氧化脂质含量较高,推测宁夏密点麻蜥可能通过促进能量代谢、机体新陈代谢等作用发挥解毒排毒、修复损伤作用。Objective To detect the metabolic products of the single component of the local Ningxia medicinal material,Phrynoceph versicolor,and to screen for metabolic substances related to disease treatment.Methods The T3 method chromatography acquisition conditions and non-targeted metabolomics technology were used detect the mixtures of artificially bred Phrynocephalus versicolor(dry samples).Ultra Performance Liquid Chromatography(UPLC)and Tand Mass Spectrometry(MS/MS)technologies were used to identify the metabolites in the samples,and the metabolite results were analyzed in combination with the.Results The mass spectrum of the single-component alcohol extract of Phrynocephalus versicolor was compared with the mass spectra of various substances.According to the classification of substances,43 classes of secondary substances were identified,including organic acids and their derivatives,amino acids and their derivatives,peptides,oxidized lip,nucleotides and their metabolites,acyl carnitines,amino acids,lysophosphatidylcholine,free fatty acids,lysosphatidylethanolamine,amines,benzene and its derivatives,coenzymes and vitamins,phenolic acids,sugars,indoles and derivatives,polyamines,pteridines and their derivatives,hormones and hormone-related substances,esters,monoglycerides,bile acids,phenols sugar alcohols,alcohols,others,sugar acids,phosphosugars,heterocyclic compounds,sulfonic acids,ketones,ch compounds,phosphates,sugar derivatives,sphingomyelin,aldehydes,tryptophan,hydrocarbon derivatives,diglycerides,phosphatylcholine,phosphatidylinositol,phosphatidylethanolamine,pigments,etc.The top three are organic acids and their(108),amino acids and their derivatives(98),peptides(83),and oxidized lipids(83).Conclusion The metabolites ofrynocephalus versicolor are rich in amino acids and their derivatives,organic acids and their derivatives,peptides,and oxidized lipids,which may play role in detoxification,repair of damage,and promotion of energy metabolism and body metabolism.
