摘  要:目的:探讨黄芩与黄精联合治疗对肺癌细胞(H1299细胞)增殖和凋亡的影响,以及可能发生的生物学机制。方法:用细胞计数试剂盒(CCK-8)法研究黄芩+黄精在抑制H1299肺癌细胞增殖方面的作用。用定量聚合酶链式反应或蛋白免疫印迹分析检测对氧磷酶3(PON 3),磷脂酰肌醇3-激酶(PI3K),蛋白激酶B(AKT),B淋巴细胞瘤-2基因(Bcl2)及Bcl-2同化X蛋白(Bax)的表达水平。此外,通过流式细胞术检测双黄诱导H1299细胞凋亡的作用以及线粒体膜电位和活性氧(ROS)含量的变化。结果:研究发现,将2种药物联合应用可以显著降低H1299细胞的生长,而且这种效应与药物浓度成正比(P<0.0001)。同时,双黄能有效促进H1299细胞凋亡(P<0.05,P<0.0001),增加ROS含量(P<0.01),同时损坏线粒体膜电位(P<0.05,P<0.0001),显著升高肺癌H1299细胞中Bax,p-PI3K,p-AKT的蛋白表达水平,显著降低PON 3及Bcl2的蛋白表达水平。结论:黄精与黄芩联用可通过下调PON 3表达抑制肺癌细胞增殖;且可通过PI3K/AKT通路诱导肺癌细胞线粒体途径凋亡。Objective:To investigate the effect of the combined treatment of Polygonati Rhizoma and Scutellariae Radix on the proliferation and apoptosis of lung cancer cells(H1299 cells),and the potential biological mechanisms involved.Methods:The cell counting kit-8(CCK-8)assay was used to study the effect of Polygonati Rhizoma and Scutellariae Radix on the inhibition of H1299 lung cancer cell proliferation.Quantitative polymerase chain reaction(qPCR)and Western blot were used to analyze the expression levels of paraoxonase 3(PON 3),phosphoinositide 3-kinase(PI3K),protein kinase B(AKT),B-cell lymphoma 2(Bcl2)gene,and Bcl-2-associated X protein(Bax).Additionally,flow cytometry was used to examine the induction of apoptosis by Polygonati Rhizoma and Scutellariae Radix in H1299 cells,as well as changes in mitochondrial membrane potential and reactive oxygen species(ROS)levels.Results:The study found that the combined application of the two drugs significantly inhibited the growth of H1299 cells,with this effect being dose-dependent(P<0.0001).At the same time,the combination effectively promoted apoptosis of H1299 cells(P<0.05,P<0.0001),increased ROS levels(P<0.01),and damaged the mitochondrial membrane potential(P<0.05,P<0.0001),significantly elevated expression levels of Bax,p-PI3K,and p-AKT proteins in H1299 cells,and significantly reduced the expression of PON 3 and Bcl2 proteins.Conclusion:The combined use of Polygonati Rhizoma and Scutellariae Radix inhibits lung cancer cell proliferation by downregulating PON 3 expression,and induces mitochondrial pathway apoptosis in lung cancer cells through the PI3K/AKT pathway.
