基于AngII/AT1R通路探究活血通脉颗粒治疗高血压肾损害的作用机制

摘  要:目的探讨基于血管紧张素II(Ang II)/血管紧张素Ⅱ型受体(AT1R)通路,活血通脉颗粒对高血压肾损害所发挥的促进作用。方法选取12只正常大鼠纳入正常对照组(A组),将48只的高血压肾损害大鼠分为模型对照组(B组)、中药组(C组)、贝那普利组(D组)及中药+贝那普利组(E组),C组、D组及E组分别予以活血通脉颗粒剂、贝那普利混悬液及中药+贝那普利灌胃饲养,比较各组大鼠血压及肾功能指标改善情况。结果与A组对比,其他4组大鼠SBP、DBP、尿m-ALB、尿NAG及血Scr、血BUN水平均更高(P<0.05),与B组比较,C组、D组及E组大鼠SBP、DBP、尿m-ALB、尿NAG及血Scr、血BUN水平均更低(P<0.05),与D组比较,C组及E组SBP、DBP、尿m-ALB、尿NAG及血Scr、血BUN水平均更低(P<0.05),与D组比较,E组SBP、DBP、尿m-ALB、尿NAG及血Scr、血BUN水平均更低(P<0.05)。与A组对比,其他各组大鼠24hPro、Ald、AngII、AT1R、ACE水平均更高(P<0.05),与B组比较,C组、D组及E组大鼠24hPro、Ald、AngII、AT1R、ACE水平均更低(P<0.05),与D组比较,C组及E组24hPro、Ald、AngII、AT1R、ACE水平均更低(P<0.05),与D组比较,E组24hPro、Ald、AngII、AT1R、ACE水平均更低(P<0.05)。结论活血通脉颗粒可有效改善高血压肾损害,联合贝那普利能够取得更加理想的病情改善效果。Objective To explore the role of Huoxue Tongmai Granule in promoting hypertensive renal damage based on angiotensinⅡ(AngII)/angiotensinⅡreceptor(AT1R)pathway.Method Twelve normal rats were selected and included in the normal control group(Group A).Forty-eight hypertensive rats with renal damage were divided into model control group(Group B),Chinese medicine group(Group C),Benazepril group(Group D)and Chinese medicine+Benazepril group(Group E).Groups C,D and E were fed with Huoxue Tongmai Granule,Benazepril suspension and Chinese medicine+Benazepril respectively.Result Compared with group A,the levels of SBP,DBP,urine m-ALB,urine NAG,blood Scr and blood BUN in the other four groups were higher(P<0.05),while those in groups C,D and E were lower(P The levels of SBP,DBP,urinary m-ALB,urinary NAG,blood Scr and blood BUN in group C and group E were lower(P<0.05).Compared with group D,the levels of SBP,DBP,urinary m-ALB,urinary NAG,blood Scr and blood BUN in group E were lower(P<0.05).Compared with group A,the 24-hour levels of PRO,Ald,AngII,AT1R and ACE in other groups were higher(P<0.05),compared with group B,the 24-hour levels of PRO,Ald,AngII,AT1R and ACE in groups C,D and E were lower(P<0.05).The 24-hour levels of Pro,Ald,AngII,AT1R and ACE in Group C and Group E were lower(P<0.05),and compared with Group D,the 24-hour levels of Pro,Ald,AngII,AT1R and ACE in Group E were lower(P<0.05).Conclusion Huoxue Tongmai Granule can effectively improve the renal damage caused by hypertension,and combined with Benazepril can achieve more ideal improvement effect.
