摘 要:为优化柴胡甾醇的提取纯化工艺,以柴胡为研究材料,甾醇提取率为考察指标,采用超声波协同酶法提取柴胡甾醇,通过单因素和正交试验优化柴胡甾醇提取工艺,并采用大孔树脂AB-8探索柴胡甾醇的纯化条件。结果表明,柴胡甾醇提取的最佳工艺条件为乙醇浓度60%、提取时间60 min、酶浓度1%,在此条件下柴胡甾醇的提取率为2.84%。大孔树脂纯化甾醇的最佳条件是吸附剂用量为2 g,进样浓度为0.2 mg/mL,进样体积为4 mL,在此条件下柴胡甾醇的吸附率为72.27%,解吸率为80.02%,甾醇含量提高到了31.20%。此提取纯化工艺稳定可行,可为柴胡甾醇饲料添加剂的开发利用提供依据。In order to optimize the extraction and purification process of bupleurumphytosterol,using Bupleurum chinense as a research material and extraction efficiencyof phytosterol as a index,bupleurum phytosterol were extracted by ultrasonic and cellulase,theextraction process of bupleurum phytosterolwas optimized by single-factor test and orthogonaltest,the purification condition of bupleurum phytosterol was studied by macroreticular resinAB-8.The results showed that the optimum technology for the extraction process the ethanolconcentration was 60%,cellulase concentration was 1%,the extraction time was 60 min,whichresulted in a extraction efficiency of 2.84%.The optimal conditions of purification were thatoptimum adsorbent dosage was 2.0 g,the concentration of the sample solution was 0.2 mg/mL,and the volume of the sample was 4 mL.Under the optimal conditions,the adsorption rate ofbupleurum phytosterol was 72.27%,the desorption rate was 80.02%,bupleurum phytosterolcontent was increased to 31.20%.The optimum extraction and purification processwas stable and feasible.This papercould provide a theoretical basis for development andutilization of Bupleurum chinense feed additivesl.
