摘 要:目的:观察低剂量脂多糖(LPS)预处理对心肌缺血再灌注(MIR)大鼠心肌细胞炎性因子的影响。方法:将84只SD大鼠按照数字随机表法分为假手术组、模型组、维拉帕米干预组、0.1 mg/kg LPS干预组、0.5 mg/kg LPS干预组、1.0 mg/kg LPS干预组。假手术组前4 h腹腔注射等体积生理盐水,开胸处理但不进行MIR手术;模型组术前24 h腹腔注射等体积生理盐水,行MIR手术;维拉帕米干预组术前24 h腹腔注射维拉帕米(2 mg/mL,0.8 mL/kg);0.1 mg/kg LPS干预组(LPS 0.1 mg/kg)、0.5 mg/kg LPS干预组(LPS 0.5 mg/kg)、1.0 mg/kg LPS干预组(LPS 1.0 mg/kg)腹腔注射。注射完毕后,除假手术组外其余大鼠均构建MIR模型。分别于术前和术后检测动物心电图。术后72 h行心脏超声检测,观察左室射血分数(LVEF)、左室短轴缩短率(LVFS)、左室收缩末期内径(LVESD)、左室舒张末期内径(LVEDD)、左室后壁收缩期厚度(LVPWs)、左室后壁舒张期厚度(LVPWd)、左室收缩末期容积(LVESV)、左室舒张末期容积(LVEDV)变化。采用2,3,5-氯化三苯基四氮唑(TTC)染色测量心肌梗死面积;使用Image J软件分析梗死区和非梗死区并计算心肌梗死面积百分比。苏木精-伊红(HE)染色观察心肌组织病理变化。采用酶联免疫吸附法(ELISA)测定血清炎性因子γ干扰素(IFN-γ)、白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)含量。结果:心脏超声结果显示,与假手术组比较,模型组大鼠LVEF、LVFS减小,提示心脏功能受损(P<0.05);与模型组比较,维拉帕米干预组和LPS各干预组大鼠LVEF、LVFS增加,提示心功能得到改善(P<0.05)。TTC染色结果显示,与假手术组比较,模型组大鼠心肌梗死面积增加(P<0.05);与模型组比较,LPS各干预组大鼠心肌梗死面积减小(P<0.05),其中1.0 mg/kg LPS干预组心肌梗死面积显著减少(P<0.05)。HE染色结果显示,假手术组心肌纤维排列整齐,心肌细胞形态规则,横纹未见明确异常;模型组心肌纤维�Objective:To observe the effect of low dose lipopolysaccharide(LPS)pretreatment on inflammatory factors in myocardial ischemia-reperfusion(MIR)rats.Methods:Eighty-four SD rats were divided into sham operation group,model group,verapamil intervention group,0.1 mg/kg LPS intervention group,0.5 mg/kg LPS intervention group,and 1.0 mg/kg LPS intervention group according to numerical random table method.In the sham operation group,equal volume normal saline was injected intraperitoneally 4 hours before the operation,and thoracotomy was performed without MIR operation.The model group was injected with normal saline 24 h before operation and the MIR operation was performed.Verapamil intervention group was intraperitoneally injected with verapamil(2 mg/mL,0.8 mL/kg)at 24 h before surgery.The 0.1 mg/kg LPS intervention group(LPS 0.1 mg/kg),0.5 mg/kg LPS intervention group(LPS 0.5 mg/kg),and 1.0 mg/kg LPS intervention group(LPS 1.0 mg/kg)were intraperitoneally injected respectively.After injection,MIR models were established in all rats except the sham operation group.ECG was detected before and after operation.Echocardiography was performed at 72 h after surgery.Left ventricular ejection fraction(LVEF),left ventricular short axis shortening rate(LVFS),left ventricular end-systolic diameter(LVESD),left ventricular end-diastolic diameter(LVEDD),left ventricular posterior wall thickness-systolic(LVPWs),left ventricular posterior wall thickness-diastolic(LVPWd),left ventricular end-systolic volume(LVESV),and left ventricular end-diastolic volume(LVEDV)were observed.Myocardial infarction area was detected by 2,3,5-triphenyltetrazolium chloride(TTC)staining.Image J software was used to analyze percentage of myocardial infarction area.The pathological changes of myocardial tissue were detected by hematoxylin-eosin(HE)staining.Serum levels of inflammatory factor interferonγ(IFN-γ),interleukin-6(IL-6),and tumor necrosis factor-α(TNF-α)were detected by enzyme-linked immunosorbent assay(ELISA).Results:The results of cardiac ultr
