摘　　要：白细胞介素-2(IL-2)在临床上主要用于抗肿瘤、抗感染的治疗。重组白细胞介素-2最早是由大肠杆菌表达的,由于表达形式是包涵体蛋白,后续需要进行变性、复性等一系列复杂的生产工艺,周期长且操作烦琐。本研究构建一系列聚肽,将白细胞介素-2和聚肽融合白细胞介素-2在大肠杆菌BL21(DE3)中分别表达,并进行电泳检测。结果显示,聚肽融合白细胞介素-2的可溶性表达量大大提高,且具有较好的细胞活性。本研究可为实验室中快速获得有活性的可溶性蛋白提供新的途径。Interleukin-2(IL-2)is mainly used for anti-tumor and anti-infection in clinic.Recombinant interleukin-2 is first expressed by Escherichia coli.Due to its expression form is inclusion body,subsequently requires a series of complex production processes including denaturation and renaturation,which is time-consuming and operationally cumbersome.In this study,a series of polypeptides are constructed,and interleukin-2 and polypeptidefused interleukin-2 are expressed in Escherichia coli BL21(DE3),respectively,and detected by electrophoresis.The results show that the soluble expression of polypeptide-fused interleukin-2 is greatly improved,and it has good cell activity.This study provides a new avenue for the rapid acquisition of active soluble proteins in the laboratory.
