摘 要:在测序过程中,荧光因具有激光照射下的可区分性和良好的生物相容性,其修饰的核苷酸在核苷酸测序等多种实验中被广泛使用。但强光的光漂白以及碱基的荧光猝灭大大增加测序产品开发的难度。基于此,该文通过使用醚键杂烷基结构对罗丹明和箐染料进行修饰。该方法有助于提高序列的特异性,测序过程中的荧光强度增强10%~15%,测序性能提升4%~5%。为测序产品的开发提供新思路。Fluorescent modified nucleotides have been used in various experiments such as nucleotide sequencing.Fluorescence during sequencing requires both distinguishability under laser irradiation and good biocompatibility.However,the strong light bleaching and fluorescence quenching of bases greatly increase the difficulty of sequencing product development.By using ether bond heteroalkylation structures to modify fluorescent rhodamine and indigo dyes to improve sequence specificity,the optical intensity of fluorescence during sequencing can be enhanced by 10%~15%and sequencing performance can be improved by 4%~5%,providing new ideas for the development of sequencing products.
