摘 要:为了探讨加味天雄散干预SCF-SRC-AKT-FYN-PLCγ信号通路调节精原干细胞增殖的作用机制,将75只清洁级SD大鼠随机分为5组(每组15只),即对照组、模型组、低剂量组、中剂量组和高剂量组.对照组予以等剂量水溶剂灌胃处理,其余4组进行腺嘌呤灌胃建立少精子症大鼠模型.建模结束后,模型组和对照组予以等剂量水溶剂治疗,其余3组分别予以低、中、高剂量加味天雄散治疗.治疗4周后检测大鼠精子浓度和活率,RT-PCR和WB分别检测睾丸组织精原干细胞特异性标志物CDH1和SCF-SRC-AKT-FYN-PLCγ信号通路各因子mRNA和蛋白表达,酶联免疫方法检测氧化应激指标SOD和MDA表达.经加味天雄散治疗后,少精子症大鼠精子浓度和精子活率增加,睾丸组织精原干细胞特异标志物CDH1和SCF-SRC-AKT-FYN-PLCγ信号通路各因子mRNA及其蛋白表达均增加,氧化应激指标SOD表达增加并且MDA表达下降,且差异有统计学意义.本研究表明加味天雄散能够促进精原干细胞增殖,提高精子浓度和精子活率,改善氧化应激,并可能通过调节SCF-SRC-AKT-FYN-PLCγ信号通路促进精原干细胞增殖.To investigate the effect of modified Tianxiong powder on the proliferation of spermatogonial stem cells through SCF-SRC-AKT-FYN-PLCγsignaling pathway.A total of 75 clean SD rats were randomly divided into 5 groups(15 in each group),namely control group,model group,low-dose group,medium-dose group and high-dose group.The control group was given equal dose of aqueous solvent intragastric treatment,and the other 4 groups were given adenine intragastric treatment to establish the rat model of oligospermia.After modeling,the model group and control group were treated with equal dose of aqueous solvent,and the other 3 groups were treated with low,medium and high doses of modified Tianxiong powder respectively.After 4 weeks of treatment,the sperm concentration and motility of the rats were detected,the mRNA and protein expressions of CDH1 and SCF-SRC-AKT-FYN-PLCγsignaling pathway in testicular tissue were detected by PCR and WB,and the expressions of SOD and MDA were detected by enzyme-linked immunoassay.The sperm concentration and sperm motility of the oligospermia rats were increased after treatment with modified Tianxiong powder.After treatment with modified Tianxiong power,the mRNA expression and protein expression of CDH1 and SCF-SRC-AKT-FYN-PLCγsignaling pathway in testicular tissue were increased.After treatment with modified Tianxiong powder,the expression of SOD increased and MDA decreased,and the difference was statistically significant.Modified Tianxiong powder can promote spermatogonial stem cell proliferation,increase sperm concentration and sperm motility,ameliorate oxidative stress,and regulate the proliferation mechanism by SCF-SRC-AKT-FYN-PLCγsignaling pathway.
