摘 要:有机小分子的结构分析,往往超出了光谱技术的分析能力,为了充分表征其结构,单晶X射线衍射方法是可用的最强大的工具之一,可以在原子水平上提供分子的结构,然而,该方法很大程度上依赖于高质量的晶体,限制了其广泛应用。近年来,新兴的晶体海绵技术(crystalline sponge, CS)打破了单晶衍射必须应用于结晶化合物的局限性,采用了一种金属有机框架(metal organic frameworks, MOFs),将一些难结晶、含量低的小分子化合物有序包合在其空腔内,使其结构确定成为可能。综述了晶体海绵技术的发展、原理、实际操作步骤及应用范围,并讨论了该技术的相对优势和局限性,为微量化合物绝对构型的确定提供参考与借鉴。The structural analysis of small organic molecules is often beyond the analytical capabilities ofspectroscopic techniques.In order to fully characterize their structure,the single-crystal X-ray diffraction method is oneof the most powerful tools available to provide molecular structure at the atomic level.However,this method relies heavilyon high-quality crystals,which limits its wide application.In recent years,the emerging crystalline sponge technology(CS)has broken the limitation that single crystal diffraction must be applied to crystalline compounds,and adopted ametal organic frameworks(MOFs).Some small molecule compounds which were difficult to crystallize and had low contentwere encapsulated into the cavity,in order to determine the structure.The development,principle,operation procedureand application range of crystal sponge technology were reviewed,and the relative advantages and limitations of thistechnology were discussed,with the aim of provide reference for determining the absolute configuration of tracecompounds.
