摘  要:【目的】探讨长链非编码RNA胰岛素样生长因子2反义转录物(lncRNA IGF2-AS)在骨髓间充质干细胞(BMSCs)成骨分化中的作用机制。【方法】选取50只6月龄SD雌性大鼠为研究对象,采用随机数字表法分为5组,每组10只,其中A组为假手术组,B组、C组、D组通过手术切除双侧卵巢,构建去卵巢(OVX)大鼠模型(各组成功率均为100%),其余10只为对照组。OVX大鼠模型构建成功后,C组大鼠给予lncRNA IGF2-AS拟似物,D组大鼠给予lncRNA IGF2-AS抑制剂,其余各组大鼠给予等体积生理盐水,各组大鼠均常规饲养8周。比较各组大鼠饲养8周后的骨密度、体重、血尿素氮水平、股骨生物力学强度、血清指标水平、lncRNA IGF2-AS、miR-3126-5p和KLK4表达水平、BMSCs成骨分化水平。【结果】各组大鼠的体重比较,差异无统计学意义(P>0.05)。B组、D组大鼠的L 5和股骨骨密度均低于A组和对照组,C组大鼠的L 5和股骨的骨密度高于A组和对照组,B组大鼠的L 5和股骨的骨密度高于D组,差异有统计学意义(P<0.05)。B组、C组、D组大鼠的血尿素氮水平均高于A组和对照组,C组大鼠的血尿素氮水平低于B组和D组,且B组低于D组,差异均有统计学意义(P<0.05)。B组、C组、D组大鼠的股骨最大载荷、杨氏模量值均低于A组和对照组,C组大鼠的股骨最大载荷、杨氏模量值均高于B组和D组,且B组高于D组,差异均有统计学意义(P<0.05)。B组、C组、D组大鼠的血清钙(Ca)、磷(P)水平均低于A组和对照组,血清碱性磷酸酶(ALP)、Ⅰ型前胶原氨基末端肽(PⅠNP)、Ⅰ型胶原C端肽(CTX-Ⅰ)水平均高于A组和对照组,C组大鼠的血清Ca、P水平均高于B组和D组,且B组高于D组,C组大鼠的血清ALP、PⅠNP、CTX-Ⅰ水平均低于B组和D组,且B组低于D组,差异均有统计学意义(P<0.05)。A组、B组、对照组大鼠的lncRNA IGF2-AS、miR-3126-5p和KLK4水平比较,差异无统计学意义(P>0.05),C组大鼠的lncRNA IGF2-AS、miR-31【Objective】To investigate the mechanism of long non-coding RNA insulin-like growth factor 2 antisense transcript(lncRNA IGF2-AS)in the osteogenic differentiation of bone marrow mesenchymal stem cells(BMSCs).【Methods】Fifty 6-month-old SD female rats were selected as the research objects and divided into 5 groups with 10 rats in each group by random number table method.Group A was sham operation group,group B,group C and group D underwent surgical resection of both ovaries to construct the ovariectomized(OVX)rat model(the success rate of each group was 100%),and the remaining 10 were control group after successful construction.After the OVX rat model was successfully constructed,the rats in group C were given lncRNA IGF2-AS simulants,the rats in group D were given lncRNA IGF2-AS inhibitors,and the other groups were given equal volume normal saline.The rats in all groups were routinely fed for 8 weeks.Bone mineral density,body weight,blood urea nitrogen level,femur biomechanical strength,serum index level,lncRNA IGF2-AS,miR-3126-5p and KLK4 expression levels,and BMSCs osteogenic differentiation level were compared in all groups after feeding for 8 weeks.【Results】There was no significant difference in body weight among all groups(P>0.05).The bone mineral density of L 5 and femur of rats in groups B and D were lower than those in group A and the control group,the bone mineral density of L 5 and femur of rats in group C was higher than that in group A and the control group,and the bone mineral density of L 5 and femur of rats in group B was higher than that in group D,with statistical significance(P<0.05).The blood urea nitrogen levels of rats in group B,C and D were higher than those in group A and the control group,the blood urea nitrogen level of rats in group C was lower than that in group B and D,and the blood urea nitrogen level of rats in group B was lower than that in group D,all difference were statistical significance(P<0.05).The maximum femur load and Young's modulus of rats in group B,C and D we
