摘 要:目的探讨肌球蛋白MYO7A在生后不同发育阶段小鼠睾丸的表达和定位。方法采用反转录PCR(RT-PCR)检测Myo7a在小鼠不同组织的表达,实时荧光定量PCR(qPCR)和蛋白免疫印迹检测MYO7A在生后不同发育阶段小鼠睾丸的表达,免疫荧光双标染色观察MYO7A在成年小鼠睾丸生精上皮不同周期中的定位。结果RT-PCR结果显示,Myo7amRNA广泛表达于小鼠多个器官,主要在睾丸中表达;qPCR和蛋白免疫印迹结果显示,MYO7A在出生后7 d小鼠睾丸中已有表达,在21d时表达量达到高峰,随后下降,在28d时降到最低,之后又逐渐升高,在成年后(56d)趋于稳定。免疫荧光双标染色结果发现MYO7A在小鼠睾丸生精上皮各期均有表达,主要位于支持细胞参与构成的顶端和基底外质特化结构上。结论MYO7A在小鼠睾丸支持细胞中高表达,主要定位于其与精子细胞之间或支持细胞之间的顶端和基底外质特化结构上,提示其可能参与支持细胞-精子细胞之间的粘附作用,以及血睾屏障的维持与动态变化过程,本研究为进一步研究MYO7A在精子发生中的功能提供了理论基础。Objective To investigate the expression and localization of myosin MYO7A in the mouse testes at different postnatal developmental stages.Methods Reverse transcription PCR(RT-PCR)was used to detect the expression of Myo7a in various mouse tissues.Real-time quantitative PCR(qPCR)and Western blotting were performed to assess the expression of MYO7A in the testes of mice at different postnatal stages.Immunofluorescence double staining was used to observe the localization of MYO7A in the seminiferous epithelium of adult mouse testes at different stages of the spermatogenic cycle.Results RT-PCR results showed that Myo7a mRNA was widely expressed in multiple organs of the mouse,with the highest expression observed in the testes.qPCR and Western blotting results indicated that MYO7A expression was detectable in the testes of 7-day-old mice,peaked at day 21,decreased thereafter,and reached its lowest level at day 28.The expression then gradually increased and stabilized after adulthood(56 days).Immunofluorescence double staining revealed that MYO7A was expressed throughout the spermatogenic cycle in the seminiferous epithelium,primarily localized to the apical and basal membrane structures of Sertoli cells,which form specialized junctions between Sertoli cells and spermatogenic cells.Conclusion MYO7A is highly expressed in Sertoli cells of the mouse testes,with its localization mainly at the apical and basal membrane structures involved in the adhesion between Sertoli cells and spermatogenic cells.This suggests that MYO7A may play a role in Sertoli cell-sperm adhesion,as well as in the maintenance and dynamic changes of the blood-testis barrier.This study provides a theoretical foundation for further research on the role of MYO7A in spermatogenesis.
