摘　　要：目的:探讨三七片对急性软组织损伤(ASTI)大鼠的治疗途径及作用靶点。方法:以重物击打致ASTI的大鼠为研究对象,经三七片干预后,在不同时间节点通过组织损伤评分、苏木精-伊红(HE)染色观察各组大鼠软组织病理形态变化;通过酶联免疫吸附实验(ELISA)检测血清中炎症因子白细胞介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)、前列腺素E_(2)(PGE_(2))和环氧合酶-2(COX-2)的含量,考察三七片对ASTI大鼠损伤的治疗效果;通过蛋白质免疫印迹(Western blot)检测大鼠软组织中COX-2和PGE_(2)蛋白质表达水平及COX-2/前列腺素E受体2(EP2)通路相关蛋白质的表达量,探究三七片治疗ASTI大鼠的作用靶点及机制。结果:三七片能有效降低ASTI大鼠血清中的IL-1β和TNF-α含量,抑制炎症反应,起到较好的抗炎作用。对大鼠血清中COX-2、PGE_(2)的定量检测及对炎症通路中COX-2、PGE_(2)和EP2表达的半定量检测结果显示,三七片能降低大鼠血清中炎症因子COX-2含量。结论:三七片通过降低ASTI模型大鼠血清中IL-1β、TNF-α、COX-2和PGE_(2)的含量,抑制组织中COX-2、PGE_(2)及EP2表达发挥抗炎作用。Objective:To investigate the therapeutic pathways and targets of Sanqi Tablets in rats with acute soft tissue injury(ASTI).Methods:A rat model of ASTI was established by impact from a heavy object.After intervention with Sanqi Tablets,the pathological morphological changes in the soft tissues of each group were observed and scored using a tissue damage score and hematoxylin-eosin(HE)staining at different time points.The serum levels of inflammatory factors,including interleukin-1β(IL-1β),tumor necrosis factor-α(TNF-α),prostaglandin E_(2)(PGE_(2)),and cyclooxygenase-2(COX-2),were measured by ELISA to evaluate the therapeutic effects of Sanqi Tablets on ASTI.Additionally,the expression levels of COX-2 and PGE_(2) proteins,as well as COX-2/prostaglandin E_(2)(EP2)pathway-related proteins in the soft tissues,were analyzed by Western blot to explore the targets and mechanisms of Sanqi Tablets in treating ASTI.Results:Sanqi Tablets effectively reduced the levels of IL-1βand TNF-αin the serum of injured rats,inhibited the inflammatory response,and exerted significant anti-inflammatory effects.Quantitative detection of COX-2 and PGE_(2) in the serum,along with semi-quantitative detection of COX-2,PGE_(2),and EP2 expressions in the inflammatory pathway,revealed that Sanqi Tablets reduced the serum levels of the inflammatory factor COX-2 in rats.Conclusion:Sanqi Tablets exert anti-inflammatory effects by reducing the serum levels of IL-1β,TNF-α,COX-2,and PGE_(2) in ASTI model rats and inhibiting the expression of COX-2,PGE_(2),and EP2 in tissues.
