摘 要:目的观察龟鹿二仙胶加味方联合泼尼松对Duchenne肌营养不良小鼠骨骼肌损伤的改善作用。方法以同源C57小鼠为正常组,将DMD模型mdx小鼠分为模型组、泼尼松组、中西药组,每组6只,予以相应药物灌胃8周。采用四肢抓力实验、转棒实验评估小鼠运动能力;ELISA法检测肌酸激酶(creatine kinase,CK);苏木精—伊红和Masson染色法观察骨骼肌的组织形态,免疫荧光法检测dystrophin蛋白表达水平;透射电子显微镜下观察小鼠骨骼肌组织的超微结构变化。结果与正常组比较,模型组小鼠抓力值显著降低(P<0.05),转棒实验掉落时间显著缩短(P<0.05),CK水平显著升高(P<0.05),骨骼肌的病理损伤和超微结构损伤严重,dystrophin蛋白几乎无表达;与模型组比较,泼尼松组和中西药组小鼠的抓力值显著升高(P<0.05),转棒实验掉落时间显著延长(P<0.05),CK水平显著降低(P<0.05),骨骼肌的病理损伤和超微结构损伤改善,可见dystrophin蛋白表达,中西药组小鼠骨骼肌病理损伤和超微结构损伤的改善程度及dystrophin蛋白表达水平优于泼尼松组。结论龟鹿二仙胶加味方联合泼尼松可显著改善DMD模型mdx小鼠骨骼肌损伤,且疗效优于泼尼松,其机制可能与增加骨骼肌dystrophy蛋白的表达水平有关。Objective To investigate the role of Jiawei Guilu Erxian Glue combined with prednisone in improving skeletal muscle injury in mice with Duchenne muscular dystrophy(DMD).Methods The homozygous C57 mice were selected as normal group,and mdx mice with DMD were divided into model group,prednisone group,and Chinese and western medicine group,with 6 mice in each group.The corresponding drug was given by gavage for 8 weeks.The limb grip strength test and the rotarod test were used to assess motor ability;ELISA was used to measure creatine kinase(CK);HE staining and Masson staining were used to observe the histopathological changes of skeletal muscles;immunofluorescence assay was used to observe the protein expression level of dystrophin;a transmission electron microscope was used to observe the change in the ultrastructure of skeletal muscle.Results Compared with the normal group,the model group had a significant reduction in grip strength(P<0.05),a significantly shorter falling latency in the rotarod test(P<0.05),and a significant increase in CK level(P<0.05),as well as severe pathological and ultrastructural damage of skeletal muscle and the absence of dystrophin protein.Compared with the model group,the prednisone group and the Chinese and western medicine group had a significant increase in grip strength(P<0.05),a significantly longer falling latency in the rotarod test(P<0.05),and a significant reduction in CK level(P<0.05),as well as improvements in the pathological and ultrastructural damage of skeletal muscle and the presence of dystrophin protein,and compared with the prednisone group,the Chinese and western medicine group had significantly better improvements in the pathological and ultrastructural damage of skeletal muscle and the protein expression level of dystrophin.Conclusion Jiawei Guilu Erxian Glue combined with prednisone can significantly improve skeletal muscle injury in mdx mice with DMD and has a better clinical effect than prednisone,possibly by increasing the protein expression level of dystrophi
