摘 要:构建突变文库是定向进化研究中的一个关键步骤。随机突变是通过变异使DNA序列发生变化从而影响生物体性状和功能的人工诱导基因变异的方法,基于突变发生的环境和后续筛选过程,可分为体外突变和体内突变。综述了随机突变文库构建策略体外突变技术(如易错PCR、DNA改组、转座子插入和CRISPR/Cas系统等)和体内突变技术(如MAGE、OrthoRep和CasPER体系等)的优缺点和应用进展,为促进酶工程与合成生物学技术的发展和推动新药研发、生物能源生产等领域的创新提供了参考。Constructing mutation libraries is a key step in directed evolution studies.Random mutation is a method of artificially induced genetic variation that changes the DNA sequence by mutation to affect traits and functions of organisms.Based on the environment in which the mutation occurs and the subsequent screening process,random mutation can be divided into in vitro mutation and in vivo mutation.In this paper,we review the advantages,disadvantages,and applications progress of construction strategies for random mutation library,including in vitro mutation techniques such as error-prone PCR,DNA shuffling,transposon insertion,and CRISPR/Cas system,and in vivo mutation techniques such as MAGE,OrthoRep,and CasPER systems,in order to provide reference for promoting the development of enzyme engineering and synthetic biology technology,as well as promoting innovation in the fields of new drug development and bioenergy production.
