摘 要:目的采用生物信息学方法探究高胆固醇血症相关差异基因和潜在的治疗中药。方法通过基因表达综合(GEO)数据库下载高胆固醇血症的基因表达谱芯片,借助R软件分析差异基因;采用Cytoscape 3.9.1软件构建差异基因相互作用可视化网络,利用DAVID数据库对关键核心基因进行基因本体(GO)富集分析和京都基因与基因组百科全书(KEGG)通路富集分析;采用关联图谱(CMap)数据库预测有效小分子化合物,通过中药系统药理学数据库与分析平台(TCMSP)反向匹配,筛选出防治高胆固醇血症的潜在中药,并进行文献考证。结果筛选出关键差异基因108个,主要参与类固醇生物合成信号通路、磷脂酰肌醇3-激酶/蛋白激酶B(PI3K-Akt)信号通路、Rap1信号通路、剪接体信号通路、吞噬体信号通路;通过CMap数据库比对分析,获得9个化合物分子,其对应候选中药20味。结论本研究筛选出了高胆固醇血症的关键致病基因和有效小分子化合物,并匹配到了对应的高频中药,可为探索中药组方配伍及中医药治疗高胆固醇血症提供新思路。Objective To explore differential genes associated with hypercholesterolemia and identify potential therapeutic Chinese medicine through bioinformatics approaches.Methods The gene expression profile of hypercholesterolemia was downloaded from the gene expression omnibus(GEO)database,and differential genes were analyzed using R software.The differential gene interaction visualization network was constructed using Cytoscape 3.9.1 software,and the key core genes were subjected to gene ontology(GO)enrichment analysis and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analysis using the DAVID database.The effective small molecule compounds were predicted using the cMap database,and the potential Chinese herbs for the treatment of hypercholesterolemia were screened by reverse matching with the TCMSP database and literature verification.Results A total of 108 key differential genes were identified,mainly involved in the steroid biosynthesis signaling pathway,PI3K-Akt signaling pathway,Rap1 signaling pathway,spliceosome signaling pathway,and phagosome signaling pathway.9 compound molecules were obtained through cMap database comparison analysis,and their corresponding candidate Chinese herbs were 20.Conclusion This study identified the key pathogenic genes and effective small molecule compounds for hypercholesterolemia and matched them with frequently used Chinese herbs,providing new insights into exploring the formula matching and treatment of hypercholesterolemia with traditional Chinese medicine.
