摘 要:目的:探讨北方冬季特有条件下制作外周血淋巴细胞微核标本时,低渗时间、固定时间以及滴片湿度对制片效果的影响。方法:特别针对冬季低温干燥环境,采集静脉血进行微量全血培养,并设置不同低渗时间、固定时间和滴片湿度条件进行实验。结果:在北方冬季特有的环境条件下,不同低渗时间和固定时间对制片效果影响的差异不显著(P> 0.05),而纱布加湿滴片可显著提高制片质量(出现A级片,P <0.05),说明在冬季制片时保持滴片湿度对改善制片效果有显著作用。结论:在冬季制作外周血淋巴细胞微核标本时,保持制片过程中各步骤条件的一致性,尤其是调整滴片湿度,可有效改善制片效果,提高微核成片质量和分析结果的准确性。Objective:To investigate the effects of hypotonic time,fixation time and drip humidity on the preparation of peripheral blood lymphocyte micronucleus specimens under the unique conditions of northern winter.Methods:Especially for the low-temperature and dry environment in winter,venous blood was collected for micro whole blood culture,and experiments were conducted under different hypotonic time,fixed time,and drip humidity conditions.Results:Under the unique environmental conditions in winter,there was no significant difference in the effect of different hypotonic time and fixed time on the film making effect(P>0.05),while the use of gauze humidification droplets significantly improved the film making quality(resulting in A-level films,P<0.05),which indicated that maintaining droplet humidity during winter film making had a significant effect on improving the film making effect.Conclusion:When preparing peripheral blood lymphocyte micronucleus specimens in winter,maintaining consistency in the conditions of each step during the preparation process,especially adjusting the drip humidity,can effectively improve the preparation effect,enhance the quality of micronucleus slices,and improve the accuracy of analysis results.
