摘 要:目的探究人参水提物和醇提物对老年失眠模型大鼠睡眠情况、昼夜节律、下丘脑中食欲素表达的影响。方法将32只SD雄性大鼠完全随机分成空白组、模型组、人参组、艾司唑仑组,各8只。采用国际通用的D-半乳糖和对氯苯丙氨酸(PCPA)叠加水平台方式进行造模,造模成功后对各组大鼠灌胃4周,以艾司唑仑片为阳性对照药。戊巴比妥钠翻正实验检测睡眠持续时间和睡眠潜伏时间;自主活动分析仪检测昼夜节律;酶联免疫吸附法检测下丘脑中食欲素A(orexin-A,OXA)、食欲素B(orexin-B,OXB)含量;免疫组化染色检测下丘脑中OXA、OXB分泌;蛋白免疫印迹实验检测OXA、OXB蛋白表达。结果人参提取物灌胃治疗后,明显缩短模型大鼠睡眠潜伏期(P<0.01)、延长睡眠持续时间(P<0.01),减少在光照环境和黑暗环境中自主活动时间(P<0.05),降低下丘脑中OXA(P<0.05)与OXB(P<0.05)分泌水平,下调OXA(P<0.05)与OXB(P<0.01)蛋白表达。结论人参提取物可以改善老年睡眠剥夺模型大鼠睡眠质量、调节昼夜节律,其机制可能与抑制下丘脑中食欲素分泌有关。Objective To investigate the effects of ginseng water and ethanol extracts on sleep,circadian rhythm,and hypothalamic orexin expression in an elderly insomnia rat model.Methods Thirty-two male SD rats were randomly divided into four groups,and the model was established using the internationally recognized method of combining D-galactose and PCPA on a water platform.After successful modeling,rats in each group were orally administered correspondingly for four weeks,with estazolam tablets used as the positive control drug.The sleep duration and sleep latency were assessed using the pentobarbital sodium turning-over test,and the circadian rhythm was measured using an autonomic activity analyzer,while the OXA and OXB levels in the hypothalamus were determined through ELISA,the secretion of OXA and OXB in the hypothalamus was examined via immunohistochemical staining and the protein expression of OXA and OXB was analyzed by Western blotting.Results After gastric lavage with ginseng extracts,the sleep latency of the model rats was significantly shortened(P<0.01),while the sleep duration was prolonged(P<0.01).The autonomous activity time was reduced in both light and dark environments(P<0.05),and the secretion levels of OXA(P<0.05)and OXB(P<0.05)in the hypothalamus were decreased,and the protein expression of OXA(P<0.05)and OXB(P<0.01)was down-regulated.Conclusion The administration of ginseng extracts has been shown to enhance sleep quality and regulate circadian rhythm in elderly rats experiencing sleep deprivation,potentially through the inhibition of orexin secretion in the hypothalamus.
