摘 要:为揭示江西苦楝(Melia azedarach)群体的遗传变异模式和给江西苦楝资源保护与利用提供参考,采用15对SSR引物对江西10个苦楝群体进行遗传多样性分析,采用聚类分析和主成分分析,探究苦楝群体的遗传关系和结构。结果表明,共检测到111个等位基因,等位基因数和有效等位基因数均值分别为4.133和2.558,香农信息指数均值为0.968,观测杂合度和期望杂合度均值分别为0.419和0.512,多态性信息含量均值为0.534。苦楝群体具有中等遗传多样性,宜春群体遗传多样性最高,九江群体遗传多样性最低。苦楝群体具有中等遗传分化。分子方差分析表明,遗传变异主要来自群体内。UPGMA聚类分析和主成分分析结果均表明,群体间具有一定的地理结构特征,安远和信丰群体聚为一类,其他群体聚为一类。江西苦楝群体具有中等遗传多样性、中等遗传分化和较高基因流,遗传多样性基本呈从西南至东北降低的趋势。In order to reveal genetic variation patterns of Melia azedarach populations,and provide reference for protection and utilization of M.azedarach in Jiangxi,15 SSR primers were used to analyze genetic diversities of 10 M.azedarach populations.Cluster analysis and principal component analysis were used to explore genetic relationships and structures of M.azedarach populations.Results showed that a total of 111 alleles were detected.Average number of alleles and effective number of alleles were 4.133 and 2.558 respectively.Average Shannon's information index was 0.968.Average observed heterozygosity and expected heterozygosity were 0.419 and 0.512 respectively.Average polymorphic information content was 0.534.M.azedarach populations had moderate genetic diversity.Yichun population had the highest genetic diversity and Jiujiang population had the lowest genetic diversity.M.azedarach populations had moderate genetic differentiation.Molecular analysis of variance indicated that genetic variation mainly occurred within populations.UPGMA cluster analysis and principal component analysis showed that there were certain geographic structure characteristics among populations.Anyuan and Xinfeng populations belonged to a group,and other populations belonged to a group.M.azedarach populations in Jiangxi had moderate genetic diversities,moderate genetic differentiation and higher gene flows.Genetic diversity basically decreased from southwest to northeast.
