摘 要:近年来,三维(3D)细胞培养技术在骨组织再生、骨代谢和骨肿瘤等领域均有广泛应用。3D细胞支架和3D培养的干细胞为骨组织的修复和替代提供了新策略,患者来源的干细胞可以整合到3D培养系统中,为精准诊疗的开发和测试提供了体外模型。此外,3D培养获得的细胞衍生物在无细胞治疗领域也展现出了显著优越性。然而,该技术也面临着诸多挑战,传统方法难以全面解析复杂3D结构中的细胞状态,且现有技术体系多基于二维(2D)细胞培养模型设计,难以匹配3D培养并保证实验结果的准确性。此外,目前3D培养系统的培养参数难以标准化,独立实验结果的横向比较尚无妥善方法。本文旨在聚焦于3D培养技术的多元化方法、广泛应用及当前面临的关键挑战,对现有研究成果作一综述,以揭示该领域当前的研究状态及未来的发展趋势,并深入阐述了3D细胞培养技术的显著优势及在骨科领域中的应用与挑战。In recent years,three-dimensional(3D)cell culture technology has been widely applied in the fields of bone tissue regeneration,bone metabolism,and bone tumors.3D cell scaffolds and 3D-cultured stem cells have provided new strategies for the repair and replacement of bone tissue.Patient-derived stem cells can be integrated into 3D culture systems,thereby providing in vitro models for the development and testing of precision medicine and treatment.Moreover,cell derivatives obtained from 3D culture have demonstrated significant advantages in acellular therapy.However,this technology also faces numerous challenges.Conventional methods are inadequate for comprehensively analyzing the cellular states within complex 3D structures.Existing technical systems are largely designed based on two-dimensional(2D)cell culture models,struggle to match 3D culture,and ensure the accuracy of experimental results.Furthermore,the culture parameters of 3D culture systems are difficult to standardize,and there is a lack of robust methods for the horizontal comparison of independent experimental results.This article aims to focus on the diverse methodologies,widespread applications,and key challenges currently faced by 3D culture technology.It reviews existing research findings to reveal the current research status and future development trends in this field.Furthermore,it provides an in-depth discussion of the notable advantages of 3D cell culture technology and its applications and challenges in the field of orthopedics.
