超临界CO_(2) 介导下米曲霉脂肪酶催化裂殖壶藻油乙酯化反应

摘  要:乙酯化反应是藻油精深加工中的关键步骤,但作为典型的非水介质催化反应,该反应体系易分相,底物混溶性差,催化效率很低。本研究在米曲霉(Aspergillus oryzae,AO)脂肪酶催化裂殖壶藻(Schizochytrium sp.)油乙酯化反应中加入绿色助溶剂超临界CO_(2)(SC-CO_(2)),在10 MPa、45℃、水质量分数10%的条件下,乙酯化的初始反应速率从常规条件下的9.3 mg/h显著增加至17.7 mg/h,反应平衡时间从20 h缩短至12 h,脂肪酸乙酯得率从60.8%明显增加至77.5%,表明SC-CO_(2)在提高底物混溶、加速传质等方面作用显著,并且添加的水分有助于维持脂肪酶活性的稳定,进而提高了催化速率。此外SC-CO_(2)作为有机助溶剂的环保替代品,不仅避免了溶剂残留问题,而且其隔氧的特性使得终产物具备更优越的抗氧化性能,有效提升了产品品质。Ethyl esterification reaction is a crucial step in the advanced processing of algal oils.As a typical non-aqueous medium catalytic reaction,this system tends to undergo phase separation,leading to poor substrate miscibility and lower catalytic efficiency.In this study,a green co-solvent,supercritical carbon dioxide(SC-CO_(2)),was introduced into the enzymatic esterification of Schizochytrium sp.oil catalyzed by Aspergillus oryzae(AO)lipase.Under the conditions of 10 MPa,45℃,and an additional water content of 10%,the initial reaction rate of esterification significantly increased from 9.3 mg/h under conventional conditions to an 17.7 mg/h.The reaction equilibrium time was shortened from 20 hours to 12 hours,and the yield of fatty acid ethyl ester(FAEE)rose noticeably from 60.8%to 77.5%.These results indicated that SC-CO_(2) played a significant role in improving substrate miscibility and accelerating mass transfer,while the added water content contributed maintaining the stability of lipase activity,thereby enhancing the catalytic rate.Furthermore,as an environmentally friendly alternative to organic solvents,SC-CO_(2) did not only avoid solvent residue issues but also imparted superior antioxidant properties to the final product due to its oxygen-exclusion characteristics,effectively enhancing product quality.
