基于多源数据挖掘分析中药专利复方防治复发性流产的用药规律及作用机制

摘　　要：目的 基于中国专利公布公告网,通过数据挖掘、网络药理学分析防治复发性流产(RSA)中药专利复方的用药与配伍特点及核心药物组作用机制。方法 检索中国专利公布公告网,收集防治RSA的中药专利复方,借助Excel 2019、SPSS Modeler 18.0、Cytoscape 3.9.1、中医传承计算平台V3.5软件分析用药规律,筛选出核心药物组。运用网络药理学方法预测核心药物组的作用机制。结果 126个专利复方中包含319味中药,菟丝子、白术、桑寄生等为高频药物,功效以补虚药为主,性味以温、平、甘、苦为主,主归肝、肾、脾经。关联分析得出常用对药为菟丝子-续断、白术-续断等,高频药物聚类分析得到3个药物组合;综合分析得出核心药物组为菟丝子-白术-杜仲-续断。网络药理学显示,核心药物组与RSA有253个交集靶点,防治RSA的主要成分为异鼠李素、槲皮素等,可能通过调控PI3K-Akt、HIF-1、AGE-RAGE等多条信号通路发挥作用,Akt1、BCL2、EFGR等为关键靶点。结论 中药专利复方防治RSA主要治法为补肾健脾、益气养血、调理冲任,核心药物组主要通过延缓细胞凋亡、调控滋养层细胞的增殖与迁移、改善胎盘血流灌注、调控炎症反应等作用机制防治RSA。Objective To analyze the drug use,and compatibility characteristics,and the action mechanism of the core drug group in the prevention and treatment of recurrent spontaneous abortion(RSA),based on the Chinese Patent Publication and Announcement Network,data mining and network pharmacology were used.Methods Chinese Patent Announcement website was searched,Chinese medicine compound patents for treating RSA were collected.With the help of Excel 2019,SPSS Modeler 18.0,Cytoscape 3.9.1,and Traditional Chinese Medicine Inheritance Computing Platform V3.5,the pattern of medication was andyzed and the core drug combination was screened.The mechanism of action of the core drug combination was predicted using a network pharmacology approach.Results The 126 patented compound formulas encompass 319 Chinese medicines,with Cuscuta,Atractylodes macrocephala,Taxillus chinensis,etc.as the high-frequency medicines.Their primary efficacy lies in tonifying deficiencies,and their properties and flavors were predominantly warm,neutral,sweet,and bitter.These herbs primarily act on the liver,kidney,and spleen meridians.Commonly used pairs include Cuscuta-Radix Dipsaci,Atractylodes macrocephala-Radix Dipsaci,etc.Three medicine combinations were obtained by high-frequency medicine cluster analysis.Comprehensive analysis yielded a core drug combination of Cuscuta-Atractylodes macrocephala-Eucommia ulmoides-Radix Dipsaci.According to network pharmacology,the core drug Combination had 253 intersecting targets with RSA.The main components of treatment of RSA were isorhamnetin,quercetin,etc.These components may modulate the role of multiple signaling pathways such as PI3K-Akt,HIF-1,AGE-RAGE,etc.,and act on key targets such as Akt1,BCL2,EFGR,etc.Conclusion The Chinese medicine patentes compound treatment of RSA mainly tonified the kidney and strengthened the spleen through deficiency tonic,benefited qi and blood and regulated Chong Ren.The core drug combination treat RSA mainly by resolving apoptosis,regulating the proliferation and migration of
