摘 要:目的基于细菌回复突变(Ames)试验探索5种塑料包装材料(plastic packaging materials,PPMs)迁移物的致突变性。方法选取高密度聚乙烯(HDPE)、聚酯(PET)、聚丙烯(PP)、砜树脂聚合物(PPSU)和共聚聚酯(Tritan)这5种PPMs,参照GB 31604.1-2015《食品安全国家标准食品接触材料及制品迁移试验通则》进行迁移试验。随后参照GB 15193.4-2014《食品安全国家标准细菌回复突变试验》采用TA97、TA98、TA100、TA102和TA15355种菌株对5种PPMs迁移液进行Ames实验,并运用傅里叶红外光谱分析(FTIR)、气质联用法(GC-MS)对具有阳性结果的迁移液进行综合性成分分析。结果在迁移液浓缩倍数为0、3、30和300倍时,5种PPMs均未出现阳性结果,但其中PET迁移液表现出细胞毒性。对PET迁移液进行稀释后再次进行Ames试验,TA1535菌株(-S9)PET迁移液各剂量组诱发的回复突变菌落数均是溶剂对照的2倍以上,且呈现剂量—反应关系。PET不同稀释倍数与细菌回复突变菌落数呈剂量相关性,Pearson相关系数为0.934(P<0.05)。IR、GC-MS分析结果提示,PET迁移液的主要成分为2,6-二叔丁基苯酚、硬脂酸、油酸酰胺、邻苯二甲酸酯和抗氧剂1076。结论HDPE、PP、PPSU和Tritan迁移液浓缩0、3、30和300倍在体外不会诱导基因突变。PET乙酸迁移液稀释后体外细菌回复突变试验阳性,提示最严苛的迁移条件下的PET材料迁移液在体外可诱导基因突变。Objective To explore the mutagenicity of five plastic packaging materials(PPMs)migration fluids based on the Bacterial Reverse Mutation Test(Ames).Methods In this study,five types of PPMs,including high-density polyethylene(HDPE),polyester(PET),polypropylene(PP),sulfone resin polymer(PPSU)and copolyester(Tritan),were selected for conducting migration tests in accordance with GB 31604.1-2015"General Rules for Migration Test of Food Contact Materials and Products".Ames test was performed on the migration fluids of the five PPMs using five strains of TA97,TA98,TA100,TA102 and TA1535 as per GB 15193.4-2014 National Standard for Food Safety-Bacterial Recovery Mutation Test.And fourier transform infrared spectroscopy(FTIR)and gas chromatography-mass spectrometry(GC-MS)were employed for comprehensive component analysis of migration liquid with positive result.Results When the concentration ratio was 0,3,30,and 300 times,no positive result were observed for the five PPMs.However,cytotoxicity was detected in the PET migration solution.Subsequent dilution of the PET migration solution result ed in a dose-response relationship in the Ames test with TA1535 strain(-S9),where the number of reverted mutant colonies exceeded twice that of the solvent control.The Pearson correlation coefficient between different dilution times of PET and the number of reverting mutant colonies was calculated to be 0.934(P<0.05).Furthermore,IR and GC-MS analysis revealed that key components of the PET migration solution included 2,6-di-tert-butylphenol,stearic acid,oleic acid amide,phthalate and antioxidant 1076.Conclusion HDPE,PP,PPSU,and Tritan concentrations 0,3,30,and 300 times do not induce gene mutations in vitro.Ames test of dilution of PET migration solution was positive,indicating that PET migration solution could induce gene mutation in vitro under the most severe migration conditions.
