牛大力提取物对SD大鼠的致畸作用研究

摘  要:目的评价牛大力提取物对SD大鼠的致畸作用。方法采用GB 15193.14—2015食品安全国家标准中的致畸试验方法,将牛大力(提取物浓缩液)样品分为低(0.86 g/kg·bw)、中(2.58 g/kg·bw)、高(7.73 g/kg·bw)3个剂量组,于受孕的第6~15天灌胃给药,阴性对照组按同等容量灌胃给予纯净水。受孕第20天处死孕鼠,解剖并检测记录孕鼠体重、受孕率、黄体数、着床数、平均活胎数、活胎、吸收胎及死胎百分率;对胎鼠进行外观检查、骨骼检查及内脏检查,计算各项畸胎率(外观、骨骼、内脏)及总畸胎率、窝畸形发生率(外观、骨骼、内脏)及总的窝畸形发生率。结果牛大力(提取物浓缩液)各剂量组对SD孕鼠母体未见明显毒性、对胎鼠未见明显胚胎毒性和致畸性。结论在本实验条件下,未发现牛大力(提取物浓缩液)对大鼠具母体毒性、胚胎毒性和致畸作用。其对大鼠致畸试验的未观察到有害作用剂量(NOAEL)为7.73 g/kg·bw,折算成牛大力干品剂量为75.00 g/kg·bw,相当于人体推荐每日服用量的300倍。Objective To assess the teratogenic effects of Callerya speciosa Schot extract on SD rats.Methods The C.speciosa extract was carried out in accordance with the GB 15193.14-2015 National Food Safety Standard for teratogenicity test.During the 6th to 15th day of gestation,SD pregnant rats were divided into three groups,which were respectively administered with C.speciosa extract at dosages of 0.86 g/kg·bw(low dose group),2.58 g/kg·bw(medium dose group),and 7.73 g/kg·bw(high dose group).A negative control group was also included,which received the same volume of water via gavage.On the 20th day of gestation,the pregnant rats were sacrificed.The body weight,conception rate,number of corpus luteum,number of implanted fetuses,average number of live fetuses,percentage of live fetuses,resorbed fetuses and stillborn fetuses were recorded.Additionally,appearance inspection,skeletal examination,and internal assessment of the fetuses were performed.The incidence of deformities in fetuses and nests,including appearance,skeleton,and viscera,as well as the overall deformity rate of nests were examined.Results C.speciosa extract did not cause maternal toxicity,embryotoxicity,or teratogenicity in SD pregnant and fetal rats at all test doses.Conclusion The extract of C.speciosa has not been found to cause any toxicity,embryotoxicity or teratogenicity.The no observed adverse effect level(NOAEL)for C.speciosa extract in rats is 7.73 g/kg·bw.This amount is equivalent to 300 times the recommended daily intake of C.speciosa for humans.
