急性心肌梗死诊断标志物IL-17RA和IRAK3的表达及其与免疫细胞浸润相关性的生物信息学研究

摘  要:目的应用生物信息学方法探讨急性心肌梗死(AMI)诊断标志物白细胞介素17受体A(IL-17RA)和白细胞介素1受体相关激酶3(IRAK3)的表达及其与免疫细胞浸润的相关性。方法从基因表达综合数据库中下载AMI相关的表达矩阵GSE123342,筛选差异表达基因(DEGs),加权共表达网络分析输出模块基因;CIBERSORT算法评估AMI免疫细胞浸润类型;STRING和Cytoscape软件构建蛋白互作网络,筛选关键(hub)基因。R软件进行hub基因差异表达,绘制ROC曲线,分析诊断标记基因与差异免疫细胞间的相关性。结果共获得567个DEGs,AMI患者中记忆B细胞、γδT细胞和嗜酸性粒细胞数量减少,单核细胞、M0巨噬细胞和中性粒细胞数量增多。IL-17RA和IRAK3均在AMI样本中呈高表达,且具有较好的诊断效能;IL-17RA和IRAK3与单核细胞(r=0.40、0.28)、M0巨噬细胞(r=0.47、0.48)和中性粒细胞(r=0.57、0.62)呈正相关,与记忆B细胞(r=-0.27、-0.28)、γδT细胞(r=-0.23、-0.13)和嗜酸性粒细胞(r=-0.36、-0.35)呈负相关。结论IL-17RA和IRAK3可作为AMI的诊断标志物,且免疫细胞浸润在AMI的发生、发展中起重要作用。Objective To find potential diagnostic markers for acute myocardial infarction(AMI)and analyze the role of immune cells infiltration in this pathology.Methods The expression matrix GSE123342 related to AMI was downloaded from Gene Expression Omnibus database to screen the differentially expressed genes(DEGs)of AMI;module genes were output from weighted gene co-expression network analysis;CIBERSORT algorithm was used to evaluate the type of immune cell infiltration in AMI.Online websites STRING and Cytoscape software were used to construct a protein-protein interaction network and screen key(hub)genes.R software was used to analyze the differential expression of hub genes,and ROC curve was drawn to analyze the correlation of diagnostic marker genes with differential immune cells.Results A total of 567 DEGs were obtained.In patients with AMI,the number of memory B cells,γδT cells and eosinophils decreased,while the number of monocytes,M0 macrophages and neutrophils increased.Both IL-17RA and IRAK3 were highly expressed in AMI and showed good diagnostic value.IL-17RA and IRAK3 were positively correlated with monocytes(r=0.40 and 0.28,respectively),M0 macrophages(r=0.47 and 0.48,respectively),and neutrophils(r=0.57 and 0.62,respectively),respectively;while they were negatively correlated with memory B cells(r=-0.27 and-0.28,respectively),gamma delta T cells(r=-0.23 and-0.13,respectively),and eosinophils(r=-0.36 and-0.35,respectively).Conclusion IL-17RA and IRAK3 can be used as diagnostic markers of AMI,and immune cell infiltration plays an important role in the occurrence and development of AMI.
