摘 要:目的:通过网络药理学方法探索石仙桃抗癌成分的作用机制及其潜在的临床应用。方法:整合多个数据库资源,筛选并鉴定石仙桃中的5种主要活性成分,并通过SwissTargetPrediction平台预测了这些成分的作用靶点。利用GO和KEGG富集分析,阐明这些靶点在细胞生物学过程中的作用,特别是在蛋白质磷酸化、激素反应等方面的调节作用。结果:从石仙桃112种化学成分中筛选出有效成分为黄菲素、欧前胡素、槲皮素、过氧麦角甾醇和丹参酮ⅡA。富集分析结果包括生物过程4400项、细胞组分406项、分子功能826项,KEGG通路249条,PPI网络确定其101个核心靶点并识别出前12位PPI核心靶点,分别为PIK3R1、JAK2、AKT1、MAPK14、ESR1、JAK1、PTK2、PTPN11、CDC25C、ERBB2、PTPN6、CDC25A。NIMNT数据库筛选出适应证共计670个,包括肌肉骨骼系统癌症、淋巴细胞白血病、结缔组织癌等。通过“石仙桃-核心靶点-疾病”网络互作图直观看出石仙桃AKT1、ERBB2、ESR1、MMP3、PTK2、MDM2等为主要核心靶点。通过PPI网络及靶基因疾病预测,以“药物-靶标-疾病”互作图进行结果可视化。结论:石仙桃在腺癌、细胞性良性肿瘤、结缔组织癌、女性生殖器官癌、肌肉骨骼系统癌的抗癌方面可能具有较好的疗效,为石仙桃在抗癌领域的应用提供了科学依据,奠定了理论基础。Objective:To exploring the mechanisms of action and potential clinical applications of anti-cancer constituents in Pholidota chinensis Lindl.through network pharmacology.Methods:Initially,multiple database resources were integrated to screen and identify the main active constituents in Pholidota chinensis Lindl.,including physalin,osthole,quercetin,ergosterol peroxide,and tanshinone IIA,and their targets were predicted using the SwissTargetPrediction platform.Subsequently,GO and KEGG enrichment analyses were conducted to elucidate the roles of these targets in cellular biological processes,especially in protein phosphorylation and hormone response regulation.Results:By constructing a protein-protein interaction(PPI)network,the interrelationships among the core targets of these active constituents were further revealed.Additionally,potential indications for the active constituents of Pholidota chinensis Lindl.were predicted,and a“drug-target-disease”network was constructed to provide scientific evidence for the application of Pholidota chinensis Lindl.in cancer treatment.The results were visualized by“drug-target-disease”interactions through PPI network and target gene diseases prediction.Conclusion:This study preliminarily predicted that Pholidota chinensis Lindl.has good efficacy in the anticancer of adenocarcinoma,cellular benign tumours,connective tissue cancers,cancers of the female reproductive organs,and cancers of the musculoskeletal system,which lays a theoretical foundation for its application in the clinical treatment of related cancers.
