摘  要:目的:观察捏脊疗法干预对幼龄厌食大鼠厌食症的效应,并以胃窦部胃饥饿素(Ghrelin)为切入点初步探究捏脊干预厌食症的调控机制。方法:选择5周龄的健康雄性SD大鼠为研究对象,采取利血平腹腔注射(45μg/ml)建立幼龄厌食大鼠模型,分别为对照组、模型组、捏脊组。捏脊干预参数为12遍/d,上、下午各1次。通过体重、进食量、悬空拉尾抵抗时长、外观行为评分明确捏脊疗法的干预效应,利用酶联免疫吸附剂测定(ELISA)检测胃窦部胃饥饿素的表达,明确捏脊疗法对促食欲激素的调控作用。在此基础上,利用ELISA检测下丘脑中2-花生四烯酸甘油(2-AG)的表达,免疫组织化学染色观察下丘脑神经肽Y(NPY)、促阿片-黑素细胞皮质素原(POMC)、大麻素受体CB1亚型(CB1)、5’-腺苷一磷酸激活的蛋白激酶α1(AMPKα1)的表达,探究捏脊干预后下丘脑摄食中枢机制是否参与并改善摄食行为。结果:与对照组比较,模型组、捏脊组体重均降低(P<0.01),自捏脊干预7 d后,捏脊组较模型组体重增长趋势更明显;模型组及捏脊组进食量均降低(P<0.05)。与模型组比较,体重及进食量组间差异无统计学意义(P>0.05)。与对照组比较,模型组悬空拉尾抵抗力与捏脊组均时长减少(P<0.01),与模型组比较,捏脊组时长增加(P<0.01)。与对照组比较,模型组及捏脊组脾虚外观行为评分升高(P<0.01),与模型组比较,捏脊组评分差异无统计学意义(P>0.05)。与对照组比较,模型组胃窦Ghrelin的含量降低(P<0.05),捏脊组含量差异无统计学意义(P>0.05);与模型组比较,捏脊组含量升高(P<0.05)。捏脊疗法对下丘脑中的NPY肽、POMC肽、AMPK1、CB1、2-AG表达无明显调控作用。结论:捏脊干预可提高利血平诱导幼龄厌食大鼠进食量和体重,改善脾虚症状,其作用机制可能与提高胃窦Ghrelin含量有关。Objective:To observe the effects of pinching spine therapy on anorexia in young rats and to preliminarily explore the regulatory mechanisms of this intervention through gastric ghrelin expression in the gastric antrum.Methods:Five-week-old healthy male SD rats were selected as subjects based on traditional Chinese medicine theory of spleen deficiency to establish a model of anorexia in young rats using reserpine intraperitoneal injection(45μg/ml).The rats were divided into three groups:control,model,and pinching spine groups.The pinching spine intervention consisted of 12 repetitions per day,with one session in the morning and one in the afternoon.Intervention effects were assessed by monitoring body weight,food intake,tail suspension test resistance time,and behavioral scoring.Gastric ghrelin levels were measured by enzyme-linked immunosorbent assay(ELISA)to explore the regulation of appetite hormones.In addition,ELISA was used to measure the expression of 2-arachidonoylglycerol(2-AG)in the hypothalamus,and immunohistochemistry was employed to assess the expression of neuropeptide Y(NPY),pro-opiomelanocortin(POMC),cannabinoid receptor CB1,and AMP-activated protein kinaseα1(AMPKα1)to investigate whether the hypothalamic feeding center mechanisms are involved in improving feeding behavior following pinching spine intervention.Results:Compared to the control group,both the model and pinching spine groups showed a significant reduction in body weight(P<0.01).After 7 days of pinching spine intervention,the pinching spine group exhibited a more pronounced weight gain compared to the model group.Both model and pinching spine groups had a reduced food intake(P<0.05).No significant differences in body weight or food intake were observed between the model and pinching spine groups(P>0.05).In the tail suspension test,the resistance time of the model and pinching spine groups was significantly reduced compared to the control group(P<0.01),while the pinching spine group showed a significant increase in resistance time com
