摘  要:为研究NtHD9调控长柄腺毛头部发生的分子机制,鉴定了NtHD9杂合突变体(NtHD9-SM)、纯合突变体(NtHD9-DM)和NtHD9过表达突变体(NtHD9-OE),分析了突变体表型和突变体中茉莉酸(JA)合成、代谢、信号转导过程中的关键基因表达量。结果表明,NtHD9基因表达水平与JA含量和长柄腺毛头大小正相关,表现为:NtHD9-OE>WT>NtHD9-SM>NtHD9-DM。随着外源喷施MeJA浓度升高,NtHD9-SM和NtHD9-DM长柄腺毛密度和腺毛头增大,其长柄腺毛的头部形态在喷施200μmol/L MeJA时基本恢复至正常大小。NtHD9促进JA合成基因LOX1、AOS和AOC2,JA代谢基因JAR1,JA信号转导基因CYP94C1,NtJAZ家族基因NtJAZ3、NtJAZ5和NtJAZ7,NtHD-ZIPⅣ家族基因NtHD3、NtHD7、NtHD9、NtHD11和NtHD12的表达;而抑制JA信号通路相关基因WRKY62、WRKY68、NtJAZ12的表达。综上,烟草NtHD9基因可能通过JA信号通路促进烟草长柄腺毛头部的发育。To explore the regulatory network of NtHD9 involved in the formation of long glandular trichomes(LGTs),analyses were conducted on NtHD9-SM,NtHD9-DM,NtHD9-OE plants.The expression levels of genes involved in jasmonic acid(JA)synthesis,metabolism and signal transduction were examined in wild-type(WT)and NtHD9 transgenic plants.Results revealed that both the size of glandular heads in LGTs and JA content followed the same order across the four lines:NtHD9-OE>WT>NtHD9-SM>NtHD9-DM.With increasing concentrations of methyl jasmonate(MeJA),the densities and sizes of LGT glandular heads in NtHD9-SM and NtHD9-DM plants increased,with head morphology normalizing at 200μmol/L MeJA.RT-qPCR analysis revealed that NtHD9 expression was positively correlated with JA synthesis-related gene(LOX1,AOS and AOC2),JA metabolic gene(JAR1),JA signal transduction gene CYP94C1,several NtJAZ family members(NtJAZ3,NtJAZ5 and NtJAZ7),and NtHD-ZIPⅣfamily members(NtHD3,NtHD7,NtHD9,NtHD11 and NtHD12).These genes exhibited decreased expression in NtHD9-SM and NtHD9-DM plants,while increased expression in NtHD9-OE plants.Conversely,three JA signaling-related genes(WRKY62,WRKY68 and NtJAZ12)showed the opposite trend,showing increased expression in NtHD9-SM and NtHD9-DM plants,and decreased expression in NtHD9-OE plants.These results indicatethat NtHD9 positively regulates the formation of glandular head in LGTs through JA signaling pathway in tobacco.
