摘 要:目的:采用网络药理学探究升陷汤治疗慢性阻塞性肺疾病(COPD)的分子机制。方法:在TCMSP中检索并筛选黄芪、知母、柴胡、升麻、桔梗的活性成分和作用靶点,在蛋白质数据库Uniport统一靶点名称,通过Cytoscape软件构建化合物-靶点网络。在Gene Card、OMIM、TTD数据库中筛选COPD疾病靶点,并与升陷汤作用靶点交集处理,得到升陷汤与COPD共同靶点,构建PPI网络并筛选关键靶点,并对共同靶点进行GO富集和KEGG通路分析,将升陷汤核心成分与药物-疾病核心基因进行分子对接获取结合能。结果:筛选后得到有效成分黄芪17个、知母12个、柴胡14个,、升麻12个、桔梗4个;槲皮素、山柰酚、木犀草素、7-O-甲基异微凸剑叶莎醇、芒柄花素为重要性排名前5的化合物。得到药物靶点213个,COPD疾病靶点1 064个,取交集后共有升陷汤治疗COPD靶点108个。升陷汤治疗COPD潜在基因KEGG信号通路分析可知,主要包括癌症相关通路、TNF、MAPK、松弛素、NF-κB、Apelin等信号通路;GO功能分析结果主要包括炎症反应的调节、调节上皮细胞增殖、对外源性刺激的反应调控等。结论:升陷汤主要通过IL-6、TNF等靶点调控TNF、NF-κB、MAKB等信号通路控制炎症,发挥对COPD的治疗作用。Objective:The molecular mechanism of Shengxian Decoction in treating chronic obstructive pulmonary disease(COPD)was investigated by network pharmacology.Methods:The active components and target sites of Astragalus Astragalus,rhizome anemarrhenae,Radix bupleuri,rattletop and platycodon grandiflorum were searched and screened in TCMSP.The target names were unified in the protein database Uniport,and the compound target network was constructed by Cytoscape software.COPD disease targets were screened in GeneCard,OMIM and TTD databases and interacted with the target of Shengxian Decoction to obtain the common target of Shengxian Decoction and COPD.PPI network was constructed and key targets were screened,and the common target GO enrichment analysis and KEGG pathway analysis were performed.The binding energy was obtained by molecular docking of the core components of Shengxian Decoction with the core genes of drug-disease.Results:After screening,17 of the active ingredients were Astragalus,12 of the rhizome,14 of the bupleurum,12 of the cohoseed,4 of the platycodon;quercetin,kaempferol,luteolin,7-O-methylisoproxanthol and ononine were the top 5 compounds.After deduplicating drug targets,213 targets were obtained,1064 targets were obtained for COPD disease,and 108 targets were obtained for Shengxian Decoction for COPD after intersection.KEGG signaling pathway analysis of potential genes of Shengxian Decoction for COPD showed that it mainly included cancer-related pathway,TNF signaling pathway,MAPK signaling pathway,relaxin signaling pathway,NF-κB signaling pathway,apelin signaling pathway,etc.The results of GO functional analysis mainly include the regulation of inflammatory response,the regulation of epithelial cell proliferation,and the regulation of response to exogenous stimuli.Conclusion:Shengxian Decoction mainly regulates TNF,NF-κB,and MAPK signaling pathways through targeting IL-6 and TNF to exert its therapeutic effects on COPD by controlling inflammation.
