草珊瑚多糖和姜制江香薷多糖组合牙膏的消炎止血作用研究

摘  要:目的:探究含姜制江香薷多糖以及姜制江香薷多糖和草珊瑚多糖配伍牙膏的消炎止血功效。方法:通过构建大鼠牙龈炎模型,采用牙周探诊检测大鼠牙龈指数、菌斑指数和牙龈出血指数,ELISA法测定大鼠血浆中细胞因子IL-4、IL-10、IL-6、IL-1β、TNF-α含量评价多糖牙膏的抗炎作用;通过脂多糖诱导人牙龈上皮细胞炎症模型,采用CCK-8法选取最佳给药浓度,ELISA法测定细胞上清液中NO、IL-4、IL-10、IL-6、IL-1β、TNF-α含量评价其抗炎作用;通过构建体外血小板模型,测定OD值计算血小板聚集率,ELISA法测定TXB2的含量评价其止血作用。结果:与模型组比较,各牙膏对大鼠口腔出血指数、牙龈指数、菌斑指数水平均显著性减少,且Ⅱ号牙膏样品减少更明显;与模型组比较,各牙膏样品均能使抑炎因子含量增加,促炎因子含量降低,且Ⅱ号牙膏样品在提高抑炎因子IL-4和降低促炎因子IL-1β、TNF-α、NO具有更好的作用;Ⅱ号牙膏组与其他组比较血小板聚集率增高更明显,Ⅰ号牙膏组和Ⅱ号牙膏组与其他组比较TXB2含量增加更明显。结论:6种牙膏样品均具有抗炎作用,其中Ⅱ号牙膏样品(含姜制江香薷多糖和草珊瑚多糖)具有较好的抗炎作用;6种牙膏样品均能使血小板聚集率增高和TXB2含量增多,其中Ⅱ号牙膏样品与其他组有显著差异,具有较好的止血作用。Objective:To study the anti-inflammatory and hemostatic effects of ginger-containing Elsholtzia chinensis polysaccharide and ginger-containing Elsholtzia chinensis polysaccharide and grass coral polysaccharide in toothpaste.Methods:By constructing a rat gingivitis model,the bleeding index(BOP),gingival index(GI)and plaque index(PI)were detected by periodontal probe,and the plasma cytokines IL-4,IL-10,IL-6,TL-1βand TNF-αwere determined by ELISA to evaluate the anti-inflammatory effect of polysaccharide toothpaste.The human gingival epithelial cell inflammation model was induced by lipopolysaccharide.The optimal dose concentration was selected by CCK-8 method,and the contents of NO,IL-4,IL-10,IL-6,IL-1βand TNF-αin the cell supernatant were determined by ELISA method to evaluate its anti-inflammatory effect.The platelet aggregation rate was calculated by establishing the platelet model in vitro,and the content of TXB2 was determined by ELISA to evaluate its hemostatic effect.Results:Compared with the model group,the levels of BOP,GI and PI were significantly reduced by each toothpaste,and the reduction ofⅱtoothpaste sample was more obvious.Compared with the model group,all the toothpaste samples could increase the content of anti-inflammatory factor and decrease the content of pro-inflammatory factor,and the toothpaste sampleⅡhad a better effect on increasing the anti-inflammatory factor IL-4 and decreasing the pro-inflammatory factor IL-1β,TNF-αand NO.Compared with other groups,the platelet aggregation rate of groupⅡwas significantly increased,and the TXB2 content of groupⅡand groupⅠwas more significantly increased than that of other groups.Conclusion:The results of in vivo and in vitro antiinflammatory experiments showed that all the six kinds of toothpaste samples had anti-inflammatory effects,among which the toothpaste II sample(containing Ginger Elsholtzia chinensis polysaccharide and sarcandra polysaccharide)had better anti-inflammatory effects.The results of hemostatic experiment in vitro show
