摘 要:为探究高脂日粮(HFD)和十五烷酸(C15:0)对妊娠期母体肠道形态结构的影响,试验选取32只10周龄C57BL/6J雌性小鼠进行交配,观察到阴道栓后记为0.5 d,并将妊娠母鼠随机分为4组,每组8只,分别饲喂低脂日粮(LFD)、低脂日粮+十五烷酸(LFD+1%C15:0)、高脂日粮(HFD)、高脂日粮+十五烷酸(HFD+1%C15:0),饲养期18 d。当母鼠妊娠日龄达到18.5 d时收集母鼠十二指肠、空肠、回肠、结肠并进行HE染色,观察肠道形态结构。结果显示:饲喂HFD显著降低了十二指肠的隐窝深度,并提高了十二指肠绒隐比,添加C15:0显著提高了十二指肠上皮细胞厚度。在十二指肠形态学指标上,HFD饮食与C15:0无交互作用。饲喂HFD显著提高了空肠隐窝深度,并降低空肠绒隐比,添加C15:0对空肠形态结构无显著影响。在空肠形态学指标上,HFD饮食与C15:0无交互作用。饲喂HFD显著增加了回肠绒毛宽度和绒毛表面积,添加C15:0对其无显著影响。在回肠肠上皮细胞厚度指标上,HFD饮食与C15:0产生了交互作用,其中HFD组显著高于LFD组。饲喂HFD和添加C15:0分别对结肠形态无显著影响,但在结肠肠上皮细胞厚度上,HFD饮食和C15:0产生了交互作用,其中HFD组显著高于LFD组和HFD+1%C15:0组。综上所述,饲喂HFD未对妊娠期母体十二指肠和结肠形态结构产生不利影响,但有损害空肠和回肠形态结构趋势,而C15:0不能缓解HFD引起的空肠和回肠损伤趋势,但有改善十二指肠上皮屏障功能的作用,具有一定的应用价值。The aim of this study was to investigate the effects of high fat diet(HFD)and pentadecanoic acid(C15:0)on the intestinal morphology and structure of mammals during pregnancy.Thirty-two 10-week-old C57BL/6J female mice were selected for mating in the experiment,and the presence of a vaginal pessary was recorded as 0.5 d of pregnancy.Pregnant females were randomly divided into four groups of eight rats each,and were fed a low fat diet(LFD),low fat diet+pentadecanoic acid(LFD+1%C15:0),a high fat diet(HFD),and a high fat diet+pentadecanoic acid(HFD+1%C15:0)for a feeding period of 18 d.The duodenum,jejunum,ileum,and colon of female rats were collected and subjected to HE staining when the gestational age of female rats reached 18.5 d to observe the intestinal morphology and structure.The results showed that feeding HFD significantly reduced the depth of duodenal crypts and increased the duodenal villous-crypt ratio,and the addition of C15:0 significantly increased the thickness of duodenal epithelial cells.There was no interaction between the HFD diet and C15:0 on duodenal morphological indices.Feeding HFD significantly increased jejunal crypt depth and decreased jejunal villous crypt ratio,and the addition of C15:0 had no significant effect on jejunal morphological structure.There was no interaction between HFD diet and C15:0 on jejunal morphological indices.Feeding HFD significantly increased ileal villus width and villus surface area,which were not significantly affected by the addition of C15:0.The HFD diet interacted with C15:0 on the ileal intestinal epithelial cell thickness index,which was significantly higher in the HFD group than in the LFD group.Feeding HFD and addition of C15:0 had no significant effect on colonic morphology,respectively,but on colonic intestinal epithelial cell thickness,HFD diet and C15:0 produced an interaction,in which the HFD group was significantly higher than the LFD group and the HFD+1%C15:0 group.In conclusion,feeding HFD did not adversely affect maternal duodenal and colonic morp
