四类虫草诱导人胃癌细胞BGC-823衰老的转录组预测分析

摘  要:基于转录组测序技术预测四类常见虫草对人胃癌细胞BGC-823衰老的诱导。4 mg/mL蝉花虫草(CH组)、冬虫夏草(DX组)、普通蛹虫草(PT组)及功能性蛹虫草(XG组)处理人胃癌细胞BGC-82324 h,对细胞衰老信号途径产生的差异表达基因(P<0.05)进行表达模式聚类分析,利用STRING数据库构建差异表达基因的蛋白质相互作用网络,获得hub基因及其共表达网络。结果显示,衰老信号途径上,PT组与XG组的差异表达基因主要为下调表达,CH组与DX组以上调为主。4个实验组共有9个差异表达基因,均为下调表达。ATM和CCND1是4个实验组的共有hub基因,表达量均被显著抑制(P<0.05)。CH组、PT组与XG组hub基因主要参与细胞周期G1/S期转换的调控,DX组则与IL-6介导的细胞衰老有关。四类虫草可通过不同调控机制诱导人胃癌细胞BGC-823衰老的发生。Transcriptome sequencing technology was used to predict the senescence induction of four common cordyceps on human gastric cancer cell BGC-823.Human gastric cancer cells BGC-823 were treated with 4 mg/mL Cordyceps cicandae(CH group),Cordyceps sinensis(DX group),Cordyceps militaris(PT group)and functional Cordyceps militaris(XG group)for 24 h.After transcriptome sequencing,expression pattern cluster analysis was performed on differentially expressed genes(DEGs)(P<0.05)generated by cell senescence signaling pathway.The protein interaction network of DEGs was constructed using STRING database to obtain hub genes and their co-expression network.In the cellular senescence signal pathway,the DEGs in PT group and XG group were mainly down-regulated,while those in CH group and DX group were mainly up-regulated.A total of 9 DEGs were identified in the 4 experimental groups,all of which were down-regulated.ATM and CCND1 were the common hub genes in the four experimental groups,and the expression levels of ATM and CCND1 were significantly inhibited(P<0.05).The hub genes of CH group,PT group and XG group were mainly involved in the regulation of G1/S phase transition of cell cycle,and the hub genes of DX group were related to IL-6 mediated cell senescence.Four classes of Cordyceps can induce the senescence of human gastric cancer cell BGC-823 through different regulatory mechanisms.
