摘  要:以沙芥属植物和十字花科部分属植物叶片为试材,采用SSR和SRAP 2种分子标记技术,研究了沙芥属植物的亲缘关系与遗传多样性,以期为沙芥属植物应用提供参考依据。结果表明:7对引物检测到的有效等位基因数平均值、多态性位点百分比平均值、Nei′s基因多样性平均值和Shannon多态性指数平均值分别为1.2607、54.88%、0.1593和0.2644;沙芥属植物遗传分化75.18%发生在沙芥种内和斧翅沙芥种各类型内,24.82%存在于种间和斧翅沙芥种内各个类型间;SRAP扩增结果显示斧翅沙芥种内不同类型之间的有效等位基因数平均值、多态性位点比率平均值、Nei′s基因多样性平均值、Shannon多态性指数的平均值分别为1.7032、70.32%、0.2342、0.3533;斧翅沙芥种遗传分化80.18%存在于类型内,19.82%存在于不同类型间。SSR分析表明沙芥种的遗传多样性水平要低于斧翅沙芥种,斧翅沙芥种内特殊类型的遗传多样性水平最高,依次是宽翅类型、距果类型、齿冠类型和斧形类型。SRAP对沙芥属与十字花科部分属植物亲缘关系分析表明沙芥属与供试的芸薹属、萝卜属、大青属、诸葛菜属、拟南芥属的亲缘关系均较远。Taking the leaves of Pugionium Gaertn.plants and parts of brassicaceae plants as experimental materials,the genetic diversity and phylogenetic relationships of Pugionium Gaertn.were studied by using SSR and SRAP molecular marker techniques,in order to provide reference for the application Pugionium Gaertn..The results showed that the average number of effective alleles,the average percentage of polymorphic loci,the average values of Nei′s and Shannon′s detected by 7 pairs of primers were 1.2607,54.88%,0.1593and 0.2644,respectively;75.18%of genetic differentiation occurs within Pugioniumcornutum(L.)Gaertn.and within various types of Pugionium dolabratum Maxim,while 24.82%occurs between species and within various types of Pugionium dolabratum Maxim.The SRAP amplification results showed that the average number of effective alleles,the average percentage of polymorphic loci,the average values of Nei′s and Shannon′s were 1.7032,70.32%,0.2342and 0.3533,respectively;80.18%of the genetic differentiation occurs within the types and 19.82%was between types.SSR analysis showed that the genetic diversity level of the Pugionium cornutum(L.)Gaertn.was lower than that of the Pugionium dolabratum Maxim.The genetic diversity level of the special types was the highest,followed by the broadwing type,calcanthus type,crown type and axe type.SRAP analysis of the phylogenetic relationship between the Pugionium Gaertn.plants and parts of the Brassicaceae family showed that the Pugionium Gaertn.had a relatively distant phylogenetic relationship with the tested Brassica,Raphanus,Clerodendrum,Orychophragmus and Arabidopsis.
