摘 要:目的:探讨贞芪扶正颗粒对癌因性疲乏小鼠的影响。方法:将40只C57BL/6J小鼠随机分为空白组、模型组、单纯化疗组(顺铂4 mg·kg^(-1))、贞芪扶正颗粒(1.5 g·kg^(-1))+化疗(顺铂4 mg·kg^(-1))组。除空白组外,其余小鼠接种Lewis肺癌细胞。接种次日开始灌胃,第15天开始腹腔注射,持续28 d。采用负重游泳实验和旷场实验检测小鼠的疲劳程度,检测小鼠血白细胞(white blood cell, WBC)、红细胞(red blood cell, RBC)、血红蛋白(hemoglobin, Hb)水平。流式细胞术检测小鼠脾脏CD3^(+)、CD4^(+)、CD8^(+)细胞含量。结果:与空白组比较,模型组小鼠体质量增加;与模型组比较,治疗组小鼠体质量减少(P<0.05)。与空白组比较,模型组小鼠负重游泳时间缩短(P<0.001);与模型组比较,贞芪扶正颗粒+化疗组小鼠负重游泳时间延长(P<0.05)。旷场实验显示,与空白组比较,模型组小鼠活动总路程、平均速度、活动时间减少,休息时间增加(P<0.05)。与空白组比较,模型组小鼠血WBC水平升高、RBC、Hb水平降低(P<0.01)。与模型组比较,贞芪扶正颗粒+化疗组小鼠血WBC水平降低(P<0.01)。各组小鼠脾脏CD3^(+)、CD4^(+)、CD8^(+)细胞含量及CD4^(+)/CD8^(+)比值比较,差异无统计学意义(P>0.05)。结论:贞芪扶正颗粒与化疗联用可在一定程度上缓解小鼠癌因性疲乏。Objective:To investigate the effects of Zhenqi Fuzheng Granule on cancer-caused fatigue mice.Methods:Forty C57BL/6J mice were randomly divided into blank group,model group,pure chemotherapy group(cisplatin 4 mg·kg^(-1)),and Zhenqi Fuzheng Granule(1.5 g·kg^(-1))+chemotherapy(cisplatin 4 mg·kg^(-1))group.Except for the blank group,the remaining mice were inoculated with Lewis lung cancer cells.Mice were inoculated with Lewis lung cancer cells by gavage on the next day and intraperitoneally on the 15th day for 28 d.The fatigue level of mice was detected using the weighted swimming test and the open field test,and the levels of white blood cells(WBC),red blood cells(RBC),and hemoglobin(Hb)were detected.Flow cytometry was used to detect the number of CD3^(+),CD4^(+)and CD8^(+)cells in the spleen of mice.Results:Compared with the blank group,the body mass of mice in the model group increased;compared with the model group,the body mass of mice in the treatment group decreased(P<0.05).Compared with the blank group,the weight-bearing swimming time of mice in the model group was shortened(P<0.001);compared with the model group,the weight-bearing swimming time of mice in Zhenqi Fuzheng Granule+chemotherapy group was prolonged(P<0.05).The open field experiment showed that compared with the blank group,the total distance,average speed and activity time of the mice in the model group decreased,and the rest time increased(P<0.05).Compared with the blank group,mice in the model group had increased blood WBC levels,decreased RBC and Hb levels(P<0.01).Compared with the model group,the blood WBC level of mice in Zhenqi Fuzheng Granule+chemotherapy group decreased(P<0.01).Comparison of spleen CD3^(+),CD4^(+),CD8^(+)cell counts and CD4^(+)CD8^(+)ratio of mice in each group showed no statistically significant difference(P>0.05).Conclusion:The combination of Zhenqi Fuzheng Granule and chemotherapy can alleviate cancer-caused fatigue in mice to a certain extent.
