摘 要:【目的】探明玛瑙红樱桃胚败育果实发育过程中相关基因的表达差异,为筛选胚败育相关基因提供理论依据。【方法】采用Illumina高通量测序技术,对不同发育时期(膨大期、转色期、成熟期)的玛瑙红樱桃胚败育果实样品进行转录组测序及相关分析。【结果】所有玛瑙红樱桃样品的高质量序列均在98%以上,映射序列均在85%以上,Q30均大于94%;共筛选出差异表达基因6316个,其中,不同比较组共有差异表达基因1207个;注释到GO的差异表达基因参与分子功能、细胞组分、生物过程等,参与生物过程最多;注释到KEGG通路的差异表达基因分别参与环境信息处理、生物体系统、遗传信息处理、细胞过程和新陈代谢5大通路的18个分支,注释到碳水化合物代谢分支的差异表达基因较多;1806个差异表达基因预测为转录因子,共注释到57个转录因子家族,其中,bHLH家族最多。【结论】玛瑙红樱桃胚败育果实膨大期与转色期、成熟期的基因表达差异较大,转色期与成熟期基因表达差异变小,可在果实转色前筛选胚败育相关基因进行功能验证。【Objective】The expression differences in related genes during the development process of embryo abortion fruit of Manaohong Cherry were explored to provide theoretical basis for screening embryo abortion related genes.【Method】The transcriptome sequencing and correlation analysis were performed on samples of aborted cherry embryos at different developmental stages(swollen stage,color changing stage and mature stage)by using Illumina high-throughput sequencing technology.【Result】Clean reads percentages of the samples were above 98%,and mapped reads percentages were above 85%,and Q30 was above 94%.A total of 6316 differentially expressed genes were screened,among which 1207 differentially expressed genes were found in different comparison groups.The differentially expressed genes annotated to GO were involved in molecular functions,cellular components,biological processes,with the most involvement in biological processes.The differentially expressed genes annotated to the KEGG pathway were involved in 18 branches of the five major pathways,respectively environmental information processing,biological systems,genetic information processing,cellular processes and metabolism.There were more differentially expressed genes annotated to the carbohydrate metabolism branch.A total of 1806 differentially expressed genes were predicted as transcription factors,and a total of 57 transcription factor families annotated,with the bHLH family being the most abundant.【Conclusion】The gene expression differences between the swollen,color changing and mature stages of embryo abortion fruit of Manaohong Cherry are significant,while the gene expression differences between the color changing and mature stages become smaller.Therefore,embryo abortion related genes can be screened for functional verification before fruit color changing.
