摘 要:目的借助网络药理学,初步探讨麻杏苦甘汤防治急性咽喉炎的分子机制。方法基于中药系统药理学分析平台TCMSP检索并筛选麻杏苦甘汤中药物活性成分,借用Uniprot数据库获得活性成分靶点,对于TCMSP数据库未收录的中药通过HERB数据库检索其化学成分;通过GeneCards和DrugBank数据库筛选急性咽喉炎靶点,使用FUNRICH软件获得共有靶点;通过STRING数据库和Cytoscape3.9.1分析蛋白相互作用并筛选核心靶点;通过Metascape数据库对共有靶点进行GO富集分析和KEGG通路分析。结果通过TCMSP和HERB数据库筛选到麻杏苦甘汤的活效成分共170种,活性成分靶点380个,与急性咽喉炎的共有靶点118个;通过蛋白相互作用分析获得麻杏苦甘汤可能作用于急性咽喉炎的30个核心靶点及5个关键靶点AKT1、TP53、TNF、IL6、EGFR;通过GO分析获得1770条生物过程,156条分子功能和82条细胞组分;通过KEGG通路分析并筛选发现AGE-RAGE、P13K-Akt、JAK-STAT及NF-κB信号通路可能是麻杏苦甘汤发挥作用的主要信号通路。结论通过网络药理学发现麻杏苦甘汤与急性咽喉炎存在共有靶点,麻杏苦甘汤可对急性咽喉炎具有一定的防治作用,其主要靶点可能与AKT1、TP53、TNF、IL6、EGFR等炎症因子有关,并且可能通过糖代谢相关通路、P13K-Akt信号通路、JAK-STAT信号通路及NF-κB等炎症通路发挥作用,可为后期的体内外实验提供理论指导和依据。Objective To explore the molecular mechanism of Maxing Kugan decoction in the prevention and treatment of acute pharyngitis based on network pharmacology.Methods Based on the traditional Chinese medicine system pharmacology analysis platform TCMSP,the active components of Maxing Kugan decoction were searched and screened,and the active component targets were obtained by using Uniprot database.The chemical components of traditional Chinese medicine not included in TCMSP database were searched by HERB database.The targets of acute pharyngitis were screened by GeneCards and DrugBank databases,and the common targets were obtained by FUNRICH software.STRING database and Cytoscape 3.9.1 were used to analyze protein interactions and screen core targets.GO enrichment analysis and KEGG pathway analysis were performed on the common targets through the Metascape database.Results A total of 170 active components of Maxing Kugan decoction were screened by TCMSP and HERB databases,380 active component targets,and 118 common targets with acute laryngitis;through protein interaction analysis,30 core targets and 5 key targets AKT1,TP53,TNF,IL6 and EGFR of Maxing Kugan decoction might act on acute pharyngitis were obtained.Through GO analysis,1770 biological processes,156 molecular functions and 82 cellular components were obtained.Through KEGG pathway analysis and screening,it was found that AGE-RAGE,PI3K-Akt,JAK-STAT and NF-κB signaling pathways might be the main signaling pathways of Maxing Kugan decoction.Conclusion Through network pharmacology,it is found that Maxing Kugan decoction has a common target with acute laryngitis.Maxing Kugan decoction can have a certain preventive and therapeutic effect on acute laryngitis.The main target may be related to AKT1,TP53,TNF,IL6,EGFR and other inflammatory factors,and may play a role in inflammatory pathways such as glucose metabolism-related pathways,PI3K-Akt signaling pathways,JAK-STAT signaling pathways and NF-κB,which can provide theoretical guidance and basis for later in vivo a
