嗜麦芽窄食单胞菌去除铜的最佳条件及影响因素

摘  要:为去除畜禽养殖业污水中的重金属铜,从畜禽粪便中筛选出1株能耐受重金属铜的嗜麦芽窄食单胞菌,从pH、菌体投加量、初始浓度、去除时间、温度等方面研究菌体去除铜的最佳条件,及主要污染物氮、磷、氯四环素等对嗜麦芽窄食单胞菌去除铜的影响。结果表明:在pH 5、初始质量浓度100 mg/L、菌体投加量8 g/L、去除时间90 min时,Cu^(2+)去除率最高,为76.1%。畜禽污水中氨氮在<80 mg/L时去除Cu^(2+)效果良好,总磷对去除Cu^(2+)无不良影响,氯四环素质量浓度为1 mg/L时,Cu^(2+)的去除率<1%,加入氯四环素0.01 mg/L对Cu^(2+)去除率产生影响。扫描电镜和红外光谱分析表明,菌体对Cu^(2+)的去除机理表现为明显的表面吸附作用,细胞表面的羟基、胺基、酰胺基、羧基、磷酸盐等官能团都与Cu^(2+)结合。综上,嗜麦芽窄食单胞菌处理畜禽污水中的重金属铜可以作为深度处理技术,用于处理氨氮浓度<80 mg/L的畜禽污水。To remove heavy metal copper in wastewater from livestock and poultry farming,a strain of Stenotrophomonas maltophilia that can tolerate heavy metal copper was screened from livestock and poultry manure.The optimal conditions for copper removal by the bacterium were studied from pH,bacterial dosage,initial concentration,removal time and temperature.The effects of major pollutants such as nitrogen,phosphorus,and tetracycline on copper removal by Stenotrophomonas maltophilia were also studied.The results showed that at pH of 5,initial concentration of 100 mg/L,dosage of 8 g/L,and removal rate of 76.1%at removal time of 90 min were the best removal conditions.When ammonia nitrogen in livestock sewage was lower than 80 mg/L,Cu^(2+)removal was good,and total phosphorus had no adverse effect on Cu^(2+)removal.When chlortetracycline concentration was 1 mg/L,the removal rate of Cu^(2+)was less than 1%,and the addition of chlortetracycline 0.01 mg/L had an effect on the removal rate of Cu^(2+).Scanning electron microscopy(SEM)and infrared spectros copy(FTIR)analysis indicated that the removal mechanism of Cu^(2+)by the bacterium exhibited a clear surface adsorption effect,and functional groups such as hydroxyl,amino,amide,carboxyl,and phosphate on the cell surface were involved in the binding with Cu^(2+).It is concluded that the treatment of heavy metal copper in livestock and poultry sewage by Stenotrophomonas maltophilia can be used as an advanced treatment technology,suitable for processing wastewater with an ammonia nitrogen concentration of less than 80 mg/L.
