摘 要:目的建立丹参“水-脂”二元组分的经皮微透析方法,研究付罐疗法(Fu's cupping therapy,FCT)不同处理方式的在体经皮促渗特征。方法考察灌流流速、温度和药物浓度对探针回收率的影响,采用增量法与减量法探究探针膜内外渗透性的一致性,考察探针体内外稳定性;以新西兰兔为动物模型,测定FCT不同处理时间(背部留罐5、10、15 min)对丹参水溶性成分与脂溶性成分的皮肤动力学行为。结果流速越大,回收率越低;温度越高,回收率越高;在不同药物浓度下,探针回收率与损失率近似相等;且探针回收率在10 h内稳定性良好。FCT背部留罐15 min对水溶性成分具有显著促渗作用,FCT背部留罐15 min和10 min对丹参脂溶性成分经皮促渗效果明显。结论FCT可作为一种新型的物理促渗技术。OBJECTIVE To establish the method of percutaneous microdialysis of"Water-lipid"binary components of Salviae Miltiorrhizae Radix et Rhizoma,and to study the characteristics of percutaneous penetration enhancement in vivo in different processing method of Fu's cupping therapy(FCT).METHODS The influence of perfusion velocity,temperature and drug concentration on the recovery rate of the probe was investigated.The consistency of the permeability of the probe in vitro and in vivo was investigated by incremental and decrement methods.The skin kinetics of water-soluble and fat-soluble components of Salviae Miltiorrhizae Radix et Rhizoma was studied under different FCT treatment times(FCT on the back for 5,10 and 15 min).RESULTS The higher the flow rate,the lower the recovery rate;the higher the temperature,the higher the recovery rate.Under different drug concentrations,the recovery rate and the loss rate of the probe were approximately equal;and the recovery rate of the probe was stable within 10 h.FCT on the back for 15 min had a significant effect on the permeation of water-soluble components,and FCT on the back for 15,10 min had a significant effect on the transdermal permeation of the fat-soluble components of Salviae Miltiorrhizae Radix et Rhizoma.CONCLUSION The FCT technique can be used as a new physical penetration technique.
